摘要
目的 :观察小剂量外源性脂多糖 (LPS)作为预处理剂量 ,在实验性AEP重症化研究中的作用及合理性。方法 :参照文献所使用的LPS剂量 (5 0 0 μg·kg-1)对实验性急性水肿性胰腺炎 (AEP)大鼠进行干预 ,并与未干预AEP大鼠和正常大鼠进行比较 ,观察其血压、血清淀粉酶、血清及胰组织亚硝酸盐 (NO2 -)浓度、血清TNF -α和病理改变等指标的变化。结果 :①LPS的介入使动物血压降低 ,幅度达 47.0± 10 .5mmHg,但均在 10min内恢复 ;②血清淀粉酶和NO浓度、胰组织NO浓度等均未受明显影响 ;③LPS干预组的血清TNF -α水平明显升高 ;④LPS加剧了组织炎性浸润程度 ,但未使AEP大鼠发生重症化。结论 :LPS5 0 0 μg·kg-1可以作为研究实验性AEP重症化的预处理剂量。
Objective:[WT5BZ]To investigate the role and reasonableness of exogenous lipopolysaccharide (LPS) in the development of acute pancreatitis with pretreatmental dose.[WT5HZ]Methods:[WT5BZ]Arterial blood pressure,serum amylase,serum TNF-α,serum nitrite,nitrite in pancreas and pathology of pancreas of the rats with experimental acute edema pancreatitis(AEP)were examined after being pretreated with LPS(500 μg·kg -1 ).The normal rats and the AEP rats without pretreatment were as control.[WT5HZ]Results:[WT5BZ]Compared with control groups,arterial blood pressure of experimental group decreased for 47.0±10.5 mmHg and restored in 10 min.Serum TNF-α obviously increased in experimental group.LPS exacerbated inflammation infiltration,but did not affect the development of AEP.In addition,there were not any difference in serum amylase,serum nitrite and nitrite in pancreas among three groups.[WT5HZ]Conclusion:[WT5BZ]As pretreatment dosage,it is reasonable to give LPS in 500 μg·kg -1 for the research of development of AEP.
