摘要
目的探寻中药复方中风醒脑液SD大鼠含药血清对体外培养的PC-12细胞缺血再灌注损伤的保护作用。方法采用血清药理学和原代神经细胞培养的方法,用SD大鼠的含中风醒脑液血清体外培养PC-12神经细胞,用连二亚硫酸钠造成细胞缺氧损伤模型,用H2O2造成细胞氧化损伤模型,建立缺血再灌注的体外细胞模型,用酶标仪测定吸光度,检测PC-12细胞的SOD活性及MDA含量。结果中药复方中风醒脑液SD大鼠含药血清对体外培养的神经细胞缺血再灌注损伤具有保护作用。结论中风醒脑液是一种神经细胞保护剂,可以提高SOD活性、降低MDA水平,能阻断氧自由连锁反应,从而保护神经细胞。
Objective： To study the protect influence of Zhongfengxingnao （ZFXN） medicated serum on ischemia reperfusion PC-12 cell. Methods： PC-12 cells were cultured by ZFXN medicated serum,and hypoxia injury cell model was built with sodium dithionite. Oxidative damage model was built with H202. Vitro cell model with is- ehemia and reperfusion model was made. The activity of SOD and content of MDA were detected. Results： ZFXN medicated serum could protect Ischemia-reperfusion injury of nerve .cellsin extracellular culture. Conclusion： ZFXN is a kind of neuruprotective agent which can improve the activity of SOD ,and decrease the content of MDA, and blocked oxygen free radical chain reactions to protect nerve cells.
