成年小鼠脑室下区及室周器官BrdU标记细胞的对比观察

Comparison of positive BrdU-labeled cells between subventricular zone and circumventriculer organs in the adult mouse

摘要 目的:比较成年健康小鼠脑室下区及室周器官BrdU标记细胞的数量及分布,探讨生理状态下室周器官的神经生发功能。方法:成年健康BALB/c小鼠,腹腔注射BrdU(50 mg/kg),连续3 d给药。4 d后用含4%多聚甲醛的磷酸缓冲液灌注动物,取脑制备石蜡切片,免疫组织化学染色。结果:侧脑室室下区可见大量BrdU标记细胞(33.70%±1.92%),在最后区、穹窿下器及脉络丛内BrdU标记细胞分别为(2.42%±0.38%),(2.78%±0.67%)和(1.12%±0.24%),连合下器未见BrdU标记细胞。侧脑室室下区BrdU标记细胞分别与室周器官BrdU标记细胞相比较均具有显著差异(P<0.05)。最后区及穹窿下器BrdU标记细胞的数量分别与脉络丛的BrdU标记细胞的数量相比均具有显著差异(P﹤0.05)。结论:室周器官存在少量的BrdU标记细胞,这表明生理状态下成年小鼠室周器官具有一定程度的神经生发功能;但室周器官神经生发功能比室下区的神经生发功能弱的多。 Objective: To investigate neurogenesis in adult mouse circumventriculer organs at physiological status by ob- serve the distribution and quantity of BrdU-labeling cells in the subventricular zone and circumventriculer organs(CVOs) in adult mouse. Methods: The adult health BALB/c mice were injected BrdU (50 mg/kg ) intraperitoneally for three days, the mice were perfused by using 4% paraformaldehyde in phosphate-buffered saline after four days, then the brain was processed for tissue section and immunocytochemical staining. Results: A large number positive BrdU-labeled ceils were found in subventricular zone( SVZ ,33.70 ± 1.92% ). A few positive BrdU-labeled cells were found in area postrema ( AP,2.42 ± O. 38% ), subfornical organ( SFO ,2.78 ± 0.67% ), and choroid plexus( CP, 1.12 ± 0.24% ). NO positive BrdU-labeled cells were found in subcommissural organ (SCO). The percentage of BrdU-labeled cells were observably different between SVZ and CVOs (P 〈 O. 05 ). The positive BrdU-labeled cells in AP and SFO were more than that in CP. The percentage of BrdU-labeled cells were observably different between AP, SFO and CP ( P 〈 0.05 ). Conclusion:A few BrdU-labeled cells were found in CVOs of aduh mice that suggest the existence of neurogenesis in CVOs of aduh mice in the physiological state. But neurogenesis in CVOs were much weaker than that in SVZ.
