摘要
目的观察观音合剂对肺脾气虚型大鼠模型肺泡灌洗液、肠道黏膜sIgA的影响,探讨观音合剂治疗小儿反复呼吸道感染的疗效及机制。方法健康SPF级SD大鼠80只随机分为5组,即观音合剂大剂量组、观音合剂小剂量组、脾胺肽对照组、空白对照组、模型对照组,每组16只。3周造模成功后,分别灌胃观音合剂大、小剂量煎剂,脾胺肽和生理盐水,干预30 d后,评价大鼠外观行为学,称脾脏、胸腺质量,计算脾脏、胸腺指数,放射免疫法检测肺泡灌洗液、肠道黏膜sIgA的含量。结果模型对照组大鼠肺泡灌洗液、肠道黏膜表面sIgA浓度和脾脏(胸腺)质量、指数降低,与空白对照组比较差异有高度统计意义(P<0.01)。观音合剂能改善模型组外观和行为异常,提高模型大鼠肺泡灌洗液、肠道黏膜表面sIgA浓度和脾脏、胸腺质量、指数,与模型对照组比较差异有高度统计意义(P<0.01),且大剂量组更接近于空白对照组。大剂量组和脾胺肽对照组组间差异有统计意义(P<0.01或P<0.05)。小剂量组肺泡灌洗液、肠道黏膜表面sIgA、脾脏指数、胸腺质量与脾胺肽对照组比较,差异无统计意义(P>0.05),小剂量组脾脏质量、胸腺指数与脾胺肽对照组比较,差异有高度统计意义(P<0.01)。结论观音合剂能有效提高模型大鼠肺泡灌洗液、肠道黏膜sIgA的浓度和脾脏(胸腺)质量、指数,这可能是其治疗反复呼吸道感染(肺脾气虚型)的作用机制。
Objective To observe the effect of Guanyin Mixture on the sIgA of bronchoalveolar lavage fluid and intestinal mucosa of lung and spleen deficiency rat model,and investigate deeply into its mechanism of treating pediatric respiratory tract infection.Methods Eighty healthy SD rats（SPF grade） were randomly separated into five groups： normal group,model group,spleen amino peptide group,high-dose Guanyin Mixture group,low-dose Guanyin mixture group,16 rats in each group.After three weeks,the model was made successfully.The animals were respectively breeding the high-dose and small-dose group of Guanyin Mixture,spleen amino peptide and physiological saline by gavage.After 30 days of intervention,evaluating appearance and behavior,weighing spleen and thymus,calculate spleen index and thymus index.Use radioimmunoassay to measure sIgA in bronchoalveolar lavage fluid and intestinal mucosa.Results sIgA concentration of bronchoalveolar lavage fluid and intestinal mucosa,weight and the index of spleen and thymus were lower in model group than in normal group（P0.01）.The appearance and behavior were better,sIgA concentration of bronchoalveolar lavage fluid and intestinal mucosa,weight and the index of spleen and thymus were higher in Guanyin Mixturel groups than in model group（P0.01）,and the high-dose group was more like normal group.There were statistical difference between the high-dose group and spleen amino peptide group（P0.05）.There were no statistical difference between the small-dose group and spleen amino peptide group in sIgA concentration of bronchoalveolar lavage fluid and intestinal mucosa,thymus weight and spleen index（P0.05）.There were significant difference between the small-dose group and the spleen amino peptide group in spleen weight and thymus index（P0.01）.Conclusion Guanyin Mixture can effectively raise sIgA of bronchoalveolar lavage fluid and intestinal mucosal and the weight and index of spleen and thymus of model rats,which might be the mechanism that Guanyin Mixture is effective in treating repeated respiratory infection with lung and spleen deficiency syndrome.
