CDK8在宫颈癌及宫颈上皮内瘤变中表达及意义的初步研究 被引量:26

摘要 目的:探讨细胞周期依赖性蛋白激酶8(CDK8)在宫颈癌及宫颈上皮内瘤变(CIN)中的表达及在宫颈癌的发生发展中的意义。方法:应用免疫组化方法(Envision、Supervision)检测CDK8在40例宫颈癌、120例CIN及20例正常宫颈组织中的表达,比较不同组织中CDK8表达差异;检测40例宫颈癌组织中Ki-67、P53蛋白表达情况,分析CDK8、Ki-67、P53蛋白三者表达的相关性。结果:各组间CDK8表达随病变严重程度逐渐增强(P<0.05)。单因素、多因素分析显示,CDK8在宫颈癌组织中的表达与淋巴结转移有关(t=6.587,P<0.05;t=2.384,P<0.05)。宫颈癌组织中CDK8表达与Ki-67表达呈正相关(r=0.522,P<0.05),P53高表达与CDK8、Ki-67高表达有关(P<0.05)。宫颈低度病变(CINⅠ)中病变持续组与病变痊愈组CDK8表达差异无统计学意义(P>0.05);宫颈高度病变(CINⅡ、CINⅢ)中病变持续组CDK8表达强于病变痊愈组,差异有统计学意义(P<0.05)。结论:CDK8表达上调可能与宫颈癌的发生发展有一定的关系,CDK8、Ki-67、P53蛋白的表达在宫颈癌的发生中可能存在一定的协同作用。检测CDK8表达可能有助于预测宫颈高度病变的转归。 Objective:To investigate the expression of CDK8 and its significance in development of cervi- cal carcinoma and CIN. Methods:The expression of CDK8 was determined using histochemistry in 40 cervi- cal carcinoma tissues,60 cervical high-grade disease,60 cervical low-grade disease and 20 normal cervical tissues. The expression of Ki-67,P53 in 40 cervical carcinoma tissues was also determined using immunohis- tochemistry. The relationship among expression of CDK8, Ki-67, P53 was statistically analyzed. Results.. CDK8 expression increases with disease grade ( P 〈 0. 05). Through single-factor and multi-factor analysis, positive correlation wass observed between CDK8 expression and lymphoid metastasis( t =6. 587, P 〈 0.05; t =2.384, P 〈 0.05) o In cervical carcinoma tissues, P53 expression is positively correlated with CDK8 and- Ki-67 expression( P 〈0.05) o In cervical low-grade disease,there is no significant difference of CDK8 expres- sion between the healing group and the persistent group ( P 〉 0.05). While for the cervical high-grade dis- ease,CDK8 expression in the persistent group is higher than that in the healing group( P 〈0.05)o Conclu- sions. There is a positive relationship between expression of CDK8 protein and development of cervical carci- noma. Positive relationship was observed between expression of CDK8 ~Ki-67 ,P53 protein in cervical carcino- ma. CDK8 may have an important role in predicting the prognosis of cervical high-grade disease.
