摘要
目的:研究绿原酸对大鼠肠道线粒体损伤的缓解作用。方法:采用H2O2诱导大鼠肠道线粒体氧化损伤,通过测定线粒体肿胀度、膜电位、丙二醛(MDA)含量、总超氧化物歧化酶(T-SOD)和谷胱甘肽过氧化物酶(GSH-Px)的活力,以及线粒体呼吸链复合物Ⅰ、Ⅳ、Ⅴ的活性,分析绿原酸对肠道线粒体损伤的缓解效果。结果:绿原酸可显著缓解H2O2造成的线粒体肿胀和膜电位的降低。绿原酸(0.032mg/mL和0.064mg/mL)显著降低MDA的含量,绿原酸(0.064mg/mL)能显著提高T-SOD、GSH-Px及复合物Ⅰ酶的活力。结论:绿原酸通过提高线粒体呼吸链复合物酶和抗氧化酶的活力,缓解线粒体H2O2的损伤。
This study was to explore the effect of chlorogenic acid on damaged rat intestinal mitochondria. Methods: Intestinal mitochondria of rats were treated with hydrogen peroxide. The mitochondrial swelling, the membrane potential, the concentration of MDA, T-SOD and GSH-Px activity, and the enzymatic activities of mitochondrial respiratory chain complexes I, IV, and V were compared. Results: CHA significantly inhibited mitochondrial swelling caused by hydrogen peroxide and reduced the membrane potential. CHA (0.032 mg/mL and 0.064 mg/mL) significantly decreased the concentration of MDA compared to damaged group. The enzymatic activity of T-SOD, GSH-Px and complex I in CHA group (0.064 mg/mL) were significantly increased compared to damaged group. Conclusion: CHA attenuated the mitochondrial damage induced by hydrogen peroxide through increasing mitochondrial respiratory chain complex enzymes and antioxidant enzymes activity.
