牙周炎大鼠龈沟液中β防御素浓度的检测 被引量:5

摘要 目的:通过建立大鼠的牙周炎模型,检测β防御素在牙周健康大鼠和慢性牙周炎大鼠龈沟液中浓度的变化,探讨β防御素与牙周炎的关系。方法:结扎法建立慢性牙周炎大鼠(20只)模型,分别采集结扎前,结扎后第2,4,6周末4组龈沟液(gingival crevicular fluid,GCF)样本,首先用ELISA方法检测β防御素总蛋白的浓度,然后应用蛋白质印迹法分别检测β防御素1,2的蛋白表达,并做半定量分析。利用SPSS 16.0统计软件进行单因素方差分析,P<0.05为差异有统计学意义。结果:临床观察慢性牙周炎大鼠模型的形成过程,在第2周末牙龈炎形成期龈沟液β防御素蛋白与β-防御素2浓度与结扎前均明显上升(P<0.05);在第4,6周末牙周炎期,两者的表达强度又明显降低(P<0.01);β-防御素1浓度随时间未见明显改变(P>0.05)。结论:β防御素总蛋白与β-防御素2浓度在牙周炎大鼠的龈沟液(GCF)中两者的浓度显著上升后又降低,提示两者可与慢性牙周炎的病程和致病机制密切相关。 Objective: Through the establishment of rat model of periodontitis, detect the concentrations changes between periodontal healthy rats and those with chronic periodontitis gingival crevicular fluid, explore the relationship of β defensin and periodontal disease. Methods: Ligation to establish chronic periodontitis in rats (20) model, four groups of GCF (gin- gival crevicular fluid) samples were collected at 0,2,4,6 weekends, firstly through ELISA 13 defensin, total protein concen- tration was detected, then test the protein expression of β-defensin-1 and 13-defensin-2 by the application of Western blot- ting, and make semi-quantitative analysis. Using the SPSS 16.0 statistical soRware for single-factor analysis of variance and Pearson, P 〈0.05 for the difference were statistically significant. Results: Clinical observation of rat model of chronic peri- odontitis formation process, the formation of the first two weekends of gingivitis GCF β defensin protein and β-defensin-2 concentrations were significantly increased (P 〈0.05) before ligation; in the 4,6 weekends of periodontitis, however, both expression intensity were significantly lower (P 〈0.01);β-defensin-1 concentrations was not much changed over time (P〉 0.05).Conclusions: Both concentrations of 13-defensin total protein and β-defensin-2 in the rat periodontitis gingival crevicular fluid (GCF) were significantly increased previously and decreased later, suggesting that both of them are likely to be associated closely with chronic periodontitis disease and pathogenic mechanisms.
