摘要
ε-聚赖氨酸(ε-PL)作为一种新型安全高效的生物食品防腐剂,已被用于食品保鲜和防腐领域,因此研究ε-PL提取与精制方法对工业化生产ε-PL具有重要意义。为有效去除ε-PL发酵液中的色素杂质,本文考察了7种不同型号活性炭对ε-PL离子交换洗脱液的脱色效果和ε-PL吸附情况。实验结果表明,活性炭LT-720具有ε-PL吸附量小和色素去除能力强的特性。通过脱色条件的优化,确定LT-720脱色条件为:活性炭用量2%(w/v),脱色温度85℃,pH4.0,脱色时间120min。在该条件下,实现ε-PL回收率达到93.8%,色素去除率为75%;洗脱液外观颜色也由橘红色变成淡黄色。全波长扫描结果显示,LT-720能够有效去除吸收峰在300nm 700nm的色素。上述研究结果为ε-PL产品的进一步纯化提供了参考依据。
ε-Poly-L-lysine (ε-PL),a novel and safe biological food preservative,has been applied in the food industry.Therefore,it is worth to investigate technologies for ε-PL extraction and purification.To effectively remove pigment in the ε-PL fermentation broth,the effects of active charcoal obtained from different manufacturers on ion exchange process were investigated in terms of decolorization and ε-PL recovery.The results showed that activated charcoal LT-720 was the best for pigment removal and low ε-PL absorption.The optimum decolorization conditions were determined through single factor experiments as follows:activated carbon charcoal 2% (w/v),decolorization temperature 85℃,pH4.0 and decolorization time 120 min.Under these conditions,the recovery rate of e-PL and decolorization rate were 93.8% and 75%,respectively.Moreover,the decolorization solution was light yellow compared with original orange.Full wavelength scanning showed that LT-720 could effectively remove the pigment in the range of 300 nm ~700 nm.The study provides a reference for the further ε-PL purification.
