摘要
目的观察白芍总苷(TGP)对体外培养的HaCaT细胞白细胞介素(IL)-6、IL-8、IL-10、干扰素(IFN)-γ分泌的影响,以及肿瘤坏死因子(TNF)-α诱导的HaCaT细胞IL-6、IL-8、IL-10、IFN-γ分泌在白芍总苷的作用下是否发生变化,从而进一步探讨银屑病的发病机制及白芍总苷治疗银屑病的可能作用机制。方法本实验运用TNF-α诱导HaCaT细胞,并用不同浓度的TGP进行处理,通过ELISA法检测TGP作用前后细胞因子分泌的变化。结果 TGP对经TNF-α诱导的HaCaT细胞IL-6、IL-8、IFN-γ分泌有抑制作用,而对IL-10分泌有促进作用。且这两种作用均与药物作用浓度相关。结论 TGP可能通过抑制HaCaT细胞IL-6、IL-8及IFN-γ的分泌,提高IL-10的分泌,进而发挥一定的免疫调节作用,从而达到治疗银屑病的作用。
Objective To observe the effect of total glucosides of paeony (TGP) on the IL-6, IL-8, IL-10 and IFN-γ secretion of HaCaT cell induced by TNF-α in vitro and to understand whether or not the cytokine secretion of HaCaT cell can be influenced by total ghcosides of paeonia under an inflammatory condition. Methods The HaCaT cell line was induced into an inflammatory condition.The levels of IL-6,IL-8,IL-10,IFN-γ were detected by double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) before and after induced. Results The result showed that the IL-6,IL-8,IFN-γ secretion of HaCaT cell line induced by TNF-a in vitro could be inhibited by total ghcosides of paeonia. The IL-10 secretion of HaCaT cell did not changed. Conclusion TGP can affect the secretion of IL-6,IL-8,IL-10,IFN-γ,which can effects the proliferation of HaCaT cell and achieve the goals of treat to cure the psoriasis.
