胰导素样生长因子-1(IGF-1)对大鼠视神经损伤的保护作用研究 被引量:3

摘要 目的探讨胰岛素样生长因子-1(insulin-like growth factor 1,IGF-1)对视神经损伤的保护作用。方法健康Wistar大鼠180只,左眼建立视神经夹伤模型,右眼为对照眼,于损伤模型建立后分为1 h、6 h、12 h、24 h、1周、2周共6个时间点组,损伤眼的一半玻璃体内注入生理盐水,另一半注入IGF-1,Western blot检测IGF-1受体表达,流式细胞仪动态检测视网膜神经节细胞凋亡率,闪光视觉诱发电位检查视神经损伤程度。结果正常对照组无IGF-1受体阳性表达;视神经损伤后1 h、6 h、12 h、24 h、1周、2周IGF-1受体阳性表达率分别为40.25%、57.55%、76.23%、97.22%、89.33%、76.47%,12 h、24 h、1周、2周与正常对照组比较差异均有显著统计学意义(P=0.008、0.001、0.001、0.008)。视神经损伤后1 h、6 h、12 h、24 h、1周、2周视网膜神经节细胞凋亡率分别为1.4%、4.4%、5.2%、8.5%、1.9%、0.9%,1 h、6 h、12 h、24 h与正常对照相比差异均有统计学意义(P=0.023、0.014、0.010、0.008),1周、2周与正常对照组相比差异均无统计学意义(均为P>0.05)。损伤+生理盐水组与损伤+IGF-1组P100振幅与潜伏期在损伤后1 h、6 h、12 h、24 h差异均有统计学意义(均为P<0.05),伤后1周、2周差异均无统计学意义(均为P>0.05)。结论视神经损伤后视网膜IGF-1受体表达增加,外源性加入IGF-1对视神经损伤有保护和修复作用。 Objective To investigate the protective effects of inject insulin-like growth factor -1 ( IGF-1 ) on optic nerve injury. Methods One hundred and eighty healthy Wistar rats were chosen, the optic nerve injury models were established in the left eyes,and divided into 1 hour group,5 hours group,12 hours group,24 hours group, 1 week group,2 weeks group after injury, the right eyes were set as the conrol group. NS was injected into the divided into vitreous in half of injured eyes ,and IGF-1 was in- jected in another half of injured eyes,the expression of IGF-1 receptor was detected by Western blot, flow cytometry was performed to detect the apoptosis of retinal ganglion cell, and fish visual evoked potential was performed to detect the level of the optic nerve injury. Results No positive expression of IGF-1 receptor was found in control group ,the positive rate of IGF-1 receptor at 1 hour,6 hours, 12 hours,24 hours, 1 week, 2 weeks after injury were 40.25% ,57.55% ,75.23% ,97.22% ,89.33% ,75.47% ,respectively,there were significant differences between 12 hours group, 24 hours group, 1 week group,2 weeks group and control group(P = 0.008,0. 001,0. 001,0. 008). The apoptic rate of retinal ganglion cells at 1 hour,5 hours, 12 hours,24 hours, 1 week,2 weeks after injury were 1.4% ,4.4% , 5.2% , 8.5% , 1.9% , 0.9% , respectively, there were sta- tistical differences between 1 hour group ,6 hours group, 12 hours group ,24 hours group and control group (P = 0. 023,0. 014,0. 010,0. 008 ), but there was no statistical difference between 1 week group ,2 weeks group and control group( all P 〉 0.05 ). There were statistical differences in applitude and potential of P100 wave at 1 hour, 6 hours, 12 hours,24 hours after injury between injury combined with NS injected group and IGF-1 group ( all P 〈 0. 05 ), and no statistical difference at 1 week and 2 weeks ( both P 〉 0. 05 ). Conclnsion The expression of IGF-1 receptor in retina after optic nerve increases, and the exogenous injection of IGF-1 have the protective and repair effects on optic nerve injury.
