摘要
目的 了解发热伴血小板减少综合征病毒(severe fever with thrombocytopenia syndrome virus,SFTSV)感染急性期患者血清microRNAs (miRNAs)的表达谱,鉴定与病毒感染或宿主免疫应答相关miRNAs.方法 采用TaqMan低密度芯片获得血清miRNA的表达谱,分析患者与正常人miR-NA的表达差异,鉴定与病毒感染或免疫相关的miRNA,最后用实时定量RT-PCR对部分miRNA进行验证.结果 TLDA芯片结果显示,与正常人血清相比,SFTS血清表达上调超过2倍的miRNA有117个,表达下调超过2倍的miRNA有30个.在表达差异较为明显的miRNA中,病毒感染或免疫相关miRNA有23个.对6个miRNA的表达进行定量RT-PCR验证,其中miR-155,miR-126,miR-15b,miR-19b与芯片结果一致.结论 通过TaqMan低密度芯片获得了SFTS病毒感染者血清miRNA的表达谱,其中部分miRNA与病毒感染或免疫相关.
Objective To understand the serum microRNAs profiling associated with severe fever with thromboeytopenia syndrome virus (SFTSV) infection, and to identify the miRNAs related to viral infec- tion or immune responses. Methods TaqMan Low-Density Array (TLDA) was used to analyze expression levels of miRNAs in serum samples from the patients and healthy controls. Some miRNAs associated with vi- ral infection or immune responses were examined by quantitative reverse transcriptase polymerase chain reac- tion (qRT-PCR). Results Compared with the healthy controls, TLDA revealed 147 miRNAs were differenti- ally expressed (fold-change 〉 2) in patients, among which 117 miRNAs were up-regulated and 30 were down- regulated. Twenty-three of 147 differentially expressed miRNAs were related to viral infection and immune re- sponses. Six miRNAs were examined by qRT-PCR, and four miRNAs (miR-155, miR-126, miR-15b, miR- 19b) were consistent with TLDA. Conclusions Serum microRNAs profiling associated with SFISV infection was obtained by TLDA, and some miRNAs were related to viral infection or immune responses.
