酪氨酸激酶c-Kit及phospho-c-Kit(Tyr719)在上皮性卵巢癌中的表达及意义

摘要 【目的】探讨膜受体型的酪氨酸激酶c-Kit及719位酪氨酸磷酸化的c-Kit[phospho-c-Kit(Tyr719)]的表达与上皮性卵巢癌的关系。【方法】采用免疫组织化学法分别检测c-Kit、phospho-c-Kit(Tyr719)在上皮性卵巢癌(82例)及卵巢良性肿瘤(25例)、正常卵巢组织(10例)表达情况,并分析这两项指标的表达与卵巢癌临床病理特征及预后的关系。【结果】82例卵巢癌中,c-Kit阳性表达率显著高于卵巢良性病变和正常卵巢组织(P<0.05)。c-Kit的表达水平与患者的临床分期呈负相关,Ⅰ、Ⅱ期患者的c-Kit阳性率显著高于Ⅲ、Ⅳ期患者(P<0.05)。Phospho-c-Kit(Tyr719)在卵巢癌组织、卵巢良性病变和正常卵巢组织中几乎无表达。【结论】c-Kit在卵巢癌中表达明显升高提示c-Kit可能与卵巢癌的发生发展有关。在上皮性卵巢癌中,c-Kit蛋白的表达水平与患者的临床分期呈负相关,提示c-Kit蛋白可能是在卵巢癌发展的早期阶段起作用的。而phospho-c-Kit(Tyr719)在卵巢癌组织中几乎无表达,提示c-Kit蛋白在Tyr719位点磷酸化可能不是上皮性卵巢癌发生、发展的激活途径。 [Objective] This study aimed to explore the expression patterns of c-Kit and phospho-c-Kit (Tyr719) in epithelial ovarian cancer. [ Methods ] A total of 82 patients of epithelial ovarian carcer who were treated at Sun Yat-sen University Cancer Center were enrolled along with 25 patients with benign ovarian lesions and 10 normal ovarian tissues served as controls. Expression of c-Kit and phospho-c-Kit (Tyr719) was examined using immunohistochemistry. [ Results ] As compared to the control group, a significantly greater proportion of ovarian cancer tissues were positive for c-Kit (P 〈 0.05). C-Kit expression was negatively correlated with FIGO stage. The positive rate of phospho-c-Kit (Tyr719) in ovarian cancer was low and there was no significant difference between the study group and the control group. [Conclusions] The increased c-Kit expression in epithelial ovarian cancer suggests that c-Kit may play a role in carcinogenesis of ovarian cancer. C-Kit expression was negatively correlated with FIGO stage. This result suggests that c-Kit may play a role in the early stages of ovarian cancer development. The phosphorylation in Tyr719 of c-Kit protein may not be the main mechanism of c-Kit in carcinogenesis and progression of ovarian cancer.
