摘要
目的通过观察小乌桂颗粒剂含药血清对胶原诱导关节炎(CIA)大鼠成纤维滑膜细胞(CIA-FLS)体外增殖的影响,探讨其治疗类风湿关节炎(RA)的可能机制。方法采用鸡Ⅱ胶原建立CIA大鼠模型,运用关节炎指数评分法及病理切片炎症程度评分系统对其进行评价。组织块培养法体外分离培养CIA大鼠膝关节滑膜组织中CIA-FLS。实验分为空白对照组、甲氨蝶呤(MTX)组及高、中、低剂量小乌桂颗粒剂含药血清组5组,分别干预CIA-FLS24、48、72h,MTT法检测其增殖情况,ELISA检测干预前和各组细胞上清中肿瘤坏死因子-α(TNF-α)和基质金属蛋白酶-1(MMP-1)的水平。结果高、中剂量小乌桂颗粒剂含药血清组和MTX组干预CIA-FLS24、48、72h均可明显抑制其增殖,细胞培养上清中TNF-α、MMP-1水平明显降低,与干预前及空白对照组比较差异有统计学意义(P<0.05);低剂量小乌桂颗粒剂含药血清组干预CIA-FLS48、72h可明显抑制CIA-FLS增殖,细胞培养上清中TNF-α水平明显降低,其干预CIA-FLS24、48、72h细胞培养上清中MMP-1水平均明显降低,与空白对照组比较差异有统计学意义(P<0.05)。结论小乌桂颗粒剂含药血清可能通过抑制FLS增殖,并抑制FLS分泌TNF-α、MMP-1来减轻滑膜炎症反应,该过程是其发挥治疗RA的可能机制。
Objective To evaluate the effect Xiao Wu Gui granule drug-containing serum on the proliferation of CIA rats fibroblast-like synoviocytes (CIA-FLS) in vitro. Methods CIA rat model that was induced with chicken collagen type Ⅱ, uses the arthritis index score and pathological inflammation scoring system for evaluation of the degree. CIA-FLS were isolated and cultured from the synovium of CIA rats by tissue block culture method, and then respectively treated with methotrexate (MTX), different concentrations of drug-containing serum or nothing (control group) for 24,48, and 72 h, the changes in the cell proliferation were detected by MTY assay, and the changes in the cell supernatant of TNF-α and MMP-1 cytokine concentrations were detected by ELISA assay. Results The high/medium dose drug-containing serum and MTX suppressed the CIA-FLS proliferation and reduced the TNF-α and MMP-1 cytokine concentration at 24,48,and 72 h after the exposure (P〈0.05). The low dose drug-containing serum suppressed the CIA-FLS proliferation and reduced the TNF-α cytokine concentration at 48 and 72 h after the exposure (P〈0.05).The low dose drug-containing serum reduced the MMP-1 cytokine concentration at 24,48,and 72 h after the exposure (P〈0.05). Conclusion Xiao Wu Gui drug-containing serum can inhibit FLS proliferation and secreted TNF-α and MMP-1 cytokine.
