摘要
目的:通过观察非吸烟、吸烟人群牙龈炎及牙周炎牙龈组织中内皮素1(ET-1)和一氧化氮合成酶(NOS)的表达,探讨吸烟与牙周病的关系。方法:收集吸烟和非吸烟人群中正常、慢性牙龈炎及牙周炎的牙龈组织各20例,应用免疫组织化学染色及Leica IMSO图像处理系统检测其中ET-1、NOS的分布及含量变化并进行统计分析。结果:无论是非吸烟还是吸烟人群牙龈炎和牙周炎患者牙龈组织中ET-1的表达强度(OD值)均明显高于正常对照组(P<0.05),且牙周炎组明显高于牙龈炎组(P<0.05),同一牙龈状态组内非吸烟与吸烟人群相比,EF-1的表达无显著性差异(P>0.05);无论是非吸烟还是吸烟人群,牙龈炎和牙周炎患者牙龈组织中NOS的表达强度(OD值)均明显高于正常对照组(P<0.05),而牙周炎组与牙龈炎组相比无显著性差异(P>0.05),在牙周炎组中,NOS的表达强度非吸烟组明显高于吸烟组(P<0.05)。结论:正常情况下牙龈组织中存在着ET-1与NOS含量的平衡,牙周病时牙龈组织中ET-1的表达随炎症程度增加而增加,而NOS的表达不能相应上调,吸烟则进一步减少NOS的表达。
AIM:To investigate the expression of endothelin-1 (ET-1) and nitric oxide synthase (NOS) in gingival tissues of smokers with periodontal disease.METHODS:60 non-smoking and 60 smoking subjects(20 healthy,20 with gingivitis and 20 with periodontitis in each group) were included.Gingival tissues were excised and subjected to immunohistochemical staining for the detection of ET-1 and NOS.The expression of ET-1 and NOS was statistically analysed by Leica IM50 image analysis system.RESULTS:ET-1 expression in gingival tissues of gingivitis and periodontitis group was significantly higher than that of healthy groups(P < 0.05) irrespective of smoking.Furthermore,ET-1 expression was significantly higher in periodontitis group than in gingivitis group(P < 0.05).NOS expression in gingival tissues of gingivitis and periodontitis group was significantly higher than that of healthy groups (P < 0.05) irrespective of smoking.No significant difference was found between gingivitis and periodontitis groups (P > 0.05).NOS expression in non-smoking periodontis group was significantly higher than that in smoking periodontitis group (P < 0.05).CONCLUSION:A balanced expression of ET-1 and NOS exists in normal gingival tissues.Periodontal disease breaks the balance and smoking may exacerbate the imbalance,which may play a role in periodontal disease progression.
