肾上腺髓质素对内皮型一氧化氮合酶在肾肿瘤细胞中表达的影响

摘要 目的观察肾上腺髓质素(adrenomedullin,ADM)对肾肿瘤细胞中内皮型一氧化氮合酶(endothelial nitric oxide synthase,eNOS)表达的影响。方法将肾肿瘤细胞分为对照组、ADM作用组、肾上腺髓质素受体shRNA(adrenomedullin receptor shRNA,ADMR shRNA)作用组,每组又设置24、48、72h 3个时间梯度。细胞培养完成后提取细胞总蛋白,Western-Blot检测各组细胞总蛋白中eNOS的表达,观察ADM对其影响。结果 ADM作用组各时间梯度eNOS的表达均高于同时段对照组和ADMR shRNA作用组(P<0.05)。结论 ADM通过其特异性受体诱导肾肿瘤细胞中eNOS表达升高。 Objective To observe the influence of adrenomedullin (ADM)on the expression of endothelial nitric oxide synthase(eNOS) in the renal tumor cell.Methods The cell of kidney tumor was set up as control group,ADM stimulation group,ADM receptor shRNA (ADMR shRNA) stimulation group,each group had three time gradients based on 24h,48h and 72h.The total cell protein was collected after cell culture,then the expression of the eNOS was detected by Western-Blot to observe the influence by ADM.Results The expression of the eNOS in ADM stimulation group was higher than that of control group and ADMR shRNA stimulation group in all the three time gradients (P < 0.05).Conclusion ADM could induce the higher expression of the eNOS in the renal tumor cell through the specific receptor.
