地锦草水提液对移植性肝癌的抑制作用及对凋亡蛋白表达的影响 被引量:8

Influence of Herba Euphorbiae Humifusae on Auti-tumor Effect and Apoptosis Protein Expression in Transplanted Liver Cancer Mice

摘要 目的:研究地锦草对移植性肝癌H22小鼠的抑制作用及对凋亡相关蛋白Bcl-2,Bax和Caspase-3表达的影响。方法:复制小鼠移植性肝癌模型后,随机分为模型组、地锦草组和环磷酰胺组。地锦草低、中、高剂量组分别以66,132,264 mg·kg-1ig,环磷酰胺组1 mg·kg-1iv 1次,模型组和环磷酰胺组灌服同体积的蒸馏水,14 d后处死小鼠,剥离瘤组织,称肿瘤质量;比色分析法检测血清超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量;免疫组化法观察肿瘤组织Bcl-2,Bax和Caspase-3蛋白表达。结果:与模型组比较,地锦草治疗后肿瘤质量明显减轻(P<0.05);地锦草各剂量组与环磷酰胺组小鼠血清SOD均升高(P<0.05,P<0.01),血清MDA含量均下降(P<0.05,P<0.01);地锦草高、中剂量组及环磷酰胺组降低Bcl-2蛋白表达,同时提高Bax,Caspase-3蛋白表达(P<0.05),降低Bcl-2/Bax。结论:地锦草抗肿瘤机制可能是通过增强抗氧化防御系统抑制肿瘤细胞的生长,纠正机体凋亡蛋白与抗凋亡蛋白失衡,激活Caspase-3酶活性而诱导肿瘤细胞凋亡。 Objective : To study the influence of Euphorbiae Humifusae herb (EHH) on anti-tumor effect and the expression of apoptosis protein in transplanted liver cancer mice. Method: H22 liver cancer cells were inculated in the right axilla to establish solid tumor models. The animal models were randomized into five groups, the model control group, cyclophosphamide (1 mg ·kg^-1) group, the EHH (66, 132, 264 mg·kg^-1) group. The medication was given continuously for 14 days. The tumors were weighted. SOD activity and MDA content of serum were measured by the colorimetric analysis. The expression of Bcl-2, Bax and Caspase-3 in tumor tissues were determined by immunohistochemistry. Result : The size of tumor in EHH treat groups was significantly smaller than that in model control group (P 〈 0.05). EHH could significantly decrease MDA level and increase SOD level (P 〈 0.05). The expressions of Bcl-2 were decreased, while the expressions of Bax and Caspase-3 were increased in EHH high, middle dore groups (P 〈 0.05). Conclusion : EHH can inhibit the growth of liver cancer cell and its mechanism is probably related to increased antioxidation, decreased Bcl-2 expression and increased Bax and Caspase-3 expression in transplanted liver cancer mice.
