iNOS和ET-1在大鼠糖尿病早期肾组织中的动态表达及意义 被引量:7

EXPRESSION OF INDUCIBLE NITRIC OXIDE SYNTHASE AND ENDOTHENLIN-1 IN KIDNEY OF EARLY DIABETIC RATS

摘要 为探讨诱生型一氧化氮合酶 (iNOS)和内皮素 - 1(ET - 1)在糖尿病 (DM )大鼠早期肾组织中的动态表达及意义。利用链脲佐菌素 (STZ)复制糖尿病动物模型 ,免疫组化及计算机图像分析定位、半定量检测iNOS和ET - 1在肾组织中的表达。结果显示 :在DM大鼠肾组织中 ,iNOS以第 2周表达最多而第 4周表达下调 ,对照组中未见表达 ;ET - 1在正常大鼠及DM大鼠肾组织中均有表达 ,髓质多于皮质 ,DM大鼠肾小球中第1,2周较对照组明显降低 ,第 4周却升高 ,在肾小管 -间质区随病程延长而逐渐增加。由此提示NO、ET参与了DM早期肾小球高灌注和肾小管 -间质的损伤。 Objective: To clarify the expression of inducible nitric oxide synthase (iNOIS) and endothelin-1 (ET-1) in kidney of diabetic rats and their effects. Methods: Diabetes Mellitus was induced by an abdominal injection streptozotocin (55mg.kg-1 body weight). Expressions of INOS and ET-1 were examined by immunohistochemical staining and then half-guanitative analysis by Mia-2000 pathology computer image analyzer. Results: The expression of iNOS was significantly increased in different levels in diabetes groups, however; no remarkable staining was observed in normal control group: In diabetes rats, iNOS reached its peak in the 2nd week and wild decreased in the 4th week ( P <0.05, compared with the 2nd week); the positive staining of ET-1 were observed in normal control and diabetes rats; in glomeruli of diabete rats, the expression of ET-1 were decreased in the 1st, 2nd week (6.943±0.170, 6.674±0.264 vs 8.088±0.383, P <0.05), but in tubulointerstitum, ET-1 were steadly increased with the course of diabetes. Conclusions: The iNOS and ET-1 may bepathogenetic factors of early diabetic nephropathic hyperafiltration, hyperfusion and injury of diabetic tubulointerstium.
