甘肃产藏药五脉绿绒蒿有效部位对肝纤维化大鼠抗氧化系统的影响 被引量:7

摘要 目的 探讨甘肃产藏药五脉绿绒蒿有效部位对肝纤维化大鼠抗氧化系统的影响.方法 在给药的基础上,采用四氯化碳(CCl4)、喂饲高脂低蛋白复合饲料并饮用20%乙醇来复制肝纤维化大鼠模型,6w后处死大鼠,对大鼠肝脏纤维化程度评分,检测血清和肝组织各项指标,对甘肃产藏药五脉绿绒蒿各提取部位防治肝纤维化疗效进行评价.结果 复合因素肝纤维化造模结束后,模型组大鼠肝组织病理结果显示其肝纤维化处于Ⅱ~Ⅲ期;与模型组比较,甘肃产藏药五脉绿绒蒿醇提物、黄碱组以及总黄酮能显著降低大鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)与肝组织中丙二醛(MDA),明显升高肝组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT),显著改善大鼠肝纤维化的抗氧化系统;同时发现总黄酮的抗肝纤维化作用显示出一定的量效关系,而生物碱的作用不明显.结论 甘肃产藏药五脉绿绒蒿醇提物及其中的总黄酮对CCl4复合因素诱导的肝纤维化大鼠抗氧化系统有一定的保护作用. Objective To investigate the influence of active fractions of Meconopsis quintuplinervia Regel (MQR) growing in Gansu Province on antioxidant system of experimental liver fibrosis rat.Methods On the basis of drug using,model of hepatic fibrosis rat was established by using sc CCl4,eating high-fat tow-protein diet and drinking 20% alcohol.Six weeks later,the effects of the extractive parts from MQR were valuated through detecting indexes in hepatic tissue and serum.Results Extractive of 90% ethanol,total flavones and mixture of total flavones and total alkaloids from MQR growing in Gansu Province could significantly decrease ALT and AST in serum and MDA in liver tissue.They could also increase SOD and CAT in liver tissue and improve antioxidant system of experimental liver fibrosis in rat.The effects of total flavones exhibited the dose-effect relationship but the effects of total alkaloids were not obvious.Conclusions The ethanol extractive from MQR growing in Gansu Province and total flavones in it have better protective effects on antioxidant system of liver fibrosis in rats induced by CCl4 composited factor.
