摘要
目的探讨《中国药典》2010版黑顺片的薄层鉴别方法,并进行改进。方法使用单因素试验方法,考察影响黑顺片生物碱薄层层析的因素。结果以正己烷∶乙酸乙酯∶甲醇∶乙醇(5.6∶3.4∶1∶1)为展开剂,置氨蒸气饱和20min的层析缸中,在室温下展开12 cm,取出,晾干,喷以稀碘化铋钾溶液,晾干,再喷以碘化碘钾和碘化铋钾的等容混合溶液,检视,斑点清晰,分离度好。结论改进后的薄层鉴别方法效果良好,为药典的修订提供了一定的参考。
Objective To discuss the thin - layer chromatography (TLC) identification method of Heishunpian from Chinese Pharmacopoeia 2010, and make some improvements. Methods Single factor experimentation was used to investigate several influ- ential factors on TLC of the alkaloid of Heishunpian. Results N - hexane : ethyl acetate: methanol : alcohol ( 5.6 : 3.4 : 1 : 1 ) were used as developing agent together with aqua ammonia, in which the thin layer plate was saturated for 20 minutes. It was developed at room temperature for 12cm. After drying, the plate was sprayed with diluted iodized bismuth potassium, and then took out air dries, later spraying isovolumetric potassium iodine iodide and bismuth potassium iodide solution. The spots were clear and sepa- rating degree was satisfaction. Conclusion The improved method of TLC identification obtained good results; also it provided cer- tain reference for the revision of the Chinese Pharmacopoeia.
