生防青霉Penicillium striatisporum Pst10抗菌物质的分离纯化及生物活性测定 被引量:2

摘要 采用乙酸乙酯萃取、薄层层析、硅胶柱层析、高效液相色谱(HPLC)及核磁共振(NMR)等方法,对生防青霉Pst10菌株液体培养生成的抗菌物质进行分离纯化和结构鉴定,并采用液滴培养结合显微镜观察法,研究纯化的化合物对辣椒疫霉等多株供试微生物的生物活性。在HPLC分离中,获得2个含量最高的活性组分(组分1和组分4),分别将这2个组分进行1H NMR和13C NMR分析,经数据库查询,最后确定这2个组分均为Calbistrin B和Calbistrin A的混合物,组分1中Calbistrin B为主要成分,组分4中Calbistrin A为主要成分;Calbistrin B对辣椒疫霉游动孢子萌发的最低抑制浓度(MIC)为1.25μg/ml,Calbistrin B对其他供试微生物的生物活性具有明显的选择性。 Antagonistic chemicals were purified from liquid culture of strain Pst10 of PeniciUium striatiporum by ethyl acetate extraction, thin layer chromatography ( TLC), silica column chromatography and high performance liquid chromatography (HPLC) in turn. Two purified fractions were analyzed and identified by 1H NMR and 13C NMR, followed by checking corresponding database. Biological activity of purified chemicals to different microorganisms was examined by means of liquid drop culture and microscope observation. Two purified fractions (Fraction 1 and Fraction 4) were all identi- fied as mixture of Calbistrin B and Calbistrin A. Calbistrin B was the main chemical of Fraction 1, and Calbistrin A was the dominant of Fraction 4. The minimal inhibitory concentration (MIC) of Calbistrin B to zoospores germination of Phytoph- thora capsici was 1.25 μg/ml. The biological activity of Calbistrin B showed obviously selective difference to tested micro-organisms.
