清肺通络方对MPP小鼠Th17细胞信号途径调控作用的研究 被引量:13

摘要 目的探讨中药清肺通络方对支原体肺炎(MPP)小鼠Th17细胞信号途径的调控作用。方法选取BALB/c小鼠45只,随机分为正常组、模型组、阿奇霉素组、清肺通络方组和中西医结合组,以MP国际标准株(MPFH)滴鼻造模,第2天开始治疗,治疗3 d后检测肺泡灌洗液(BALF)中IL-17、IL-6、转化生长因子-β1(TGF-β1)水平,以及肺组织孤儿核受体(RORγt)、NF-κBp65蛋白定量。结果 (1)MPP小鼠BALF中IL-17、IL-6、TGF-β1水平均显著升高,清肺通络方对各指标的调节作用均优于阿奇霉素,联合应用效果最好。(2)MPP小鼠肺组织RORγt、NF-кBp65蛋白表达均增高,清肺通络方对RORγt和NF-κBp65蛋白的调节作用均优于阿奇霉素,联合应用效果最好。结论清肺通络方可调节MPP小鼠Th17细胞效应因子IL-17及其上下游转录因子IL-6、TGF-β1、RORγt、NF-κBp65。 Objective To explore the regulatory effect of Qingfei Tongluo Formula on Thl7 cell sig- nal pathway in mycoplasma pneumonia(MPP) mice. Methods Fortyfive BALB/c mice were randomized into a normal group, a model group, an Azithromycin group, a Qingfei Tongluo Formula group and an integrated Chinese and western medicine group. The model was prepared with MPFH nasal drops. Since the 2nd day, the levels of IL - 17, IL -6 and TGF - 131 were detected in BALF, as well as the expressions of ROR^t and NF - ~Bp65. Results ( 1 ) The levels of IL - 17, IL - 6 and TGF - 131 were increased significantly in BALF in MPP mice. The regulatory effect of Qingfei Tongluo Formula for all the indexes was superior to Azithromycin. The combined administration achieved the best effect. ( 2 ) The expressions of ROR',/t and NF - KBp65 of lung tissue of MPP mice were all increased. The regulatory effect of Qingfei Tongluo Formula on the protein expres- sions of ROR~/t and NF - KBp65 was superior to Azithromycin. The combined administration achieved the best effect. Conclusion Qingfei Tongluo Formula regulates the effect factor IL - 17 and the upsteam and downstream transcription factors such as IL - 6, RORt and NF - KBp65 of Thl7 cell in MPP mice.
