摘要
目的观察黄芪注射液(AI)对压疮缺血-再灌注损伤血清NO、iNOS的影响,并探讨其作用机制。方法 32只SD大鼠随机分4组,每组8只:A组(正常对照)、B组(AI腹腔注射加外涂)、C组(AI腹腔注射)和D组(缺血再灌注组),各组均采用HE染色与NO、iNOS水平测定。观察比较4组大鼠受压部位的肉眼、镜下检查以及NO、iNOS测定。结果 B组组织病理变化较C组和D组明显减轻;B组、C组和D组大鼠血清中NO、iNOS水平显著高于A组(P<0.01),B组大鼠血清中NO、iNOS水平均明显低于C组、D组(P<0.01)。结论 NO参与了压疮缺血再灌注损伤,黄芪注射液可能通过逆转NOS系统的功能紊乱,减少再灌注期NO生成,从而减轻组织损伤。黄芪注射液腹腔注射加外涂的作用优于单用黄芪注射液腹腔注射。
Objective To study the effects of Astragalus injection on the content of NO and the activity of iNOS in the pressure ul- cers involved ischemia-reperfusion (I/R) injury in rats and explore its protective mechanism. Methods 32 rats were divided into 4 groups randomly, each group with 8 rats: group A (control group); group B (AI treated group with the way of intraperitoneal injection and External use); group C (AI treated group with the way of intraperitoneal injection);group D (ischemia-reperfusion group), Each group was adopted with HE staining, Nitric oxide(NO) and nitric oxide synthase(iNOS) detection.The eye pressure, microscopic examination and NO, iNOS determination of Rats in 4 groups were observed and compared. Observation and comparisons were made among the groups.Results HE staining showed that the level of tissue pathological changes in rats of the group B was significantly decreased compared with group C and group D;The values of NO,iNOS were markedly higher in group B,C and D than those in group A(P〈 0.01).However,those in group B were significantly decreased compared with group C and D(P〈0.01).Conclusiou NO is related to is- chemia-reperfusion injury. Astragalus injection has a protective effect against tissue damage and the mechanism may be related to re- ducing the content of NO through reversing the disorder of NOS system function.The mix way of intraperitoneal injection and External use might be more effective than the way of intraperitoneal injection .
