摘要
目的探讨基于IS6110的PCR方法在唐山市结核分枝杆菌基因分型中的应用价值,分析唐山市结核分枝杆菌分离株的基因型分布特征。方法2012年1月-2013年3月从唐山市第四医院就诊的结核病患者中分离结核分枝杆菌166株,采用基于IS6110的PCR方法进行基因分型。结果唐山市结核分枝杆菌基因型以北京基因型为主导。166株结核分枝杆菌具有明显的多态性,Ⅰ型、Ⅱ型、Ⅲ型分别包含51株、88株、11株,其他型别呈散在分布。在优势基因型(Ⅱ型)中,30~60岁组高于其他年龄组;农村患者比例偏高。结论基于IS6110的PCR方法能有效进行唐山市结核分枝杆菌的基因分型,证实唐山市结核分枝杆菌呈聚集性分布。
Objective To explore the value of IS6110 - PCR fingerprinting method and analyze genotype characteristics of predominant Mycobaeterium tuberculosis among TB patients in Tangshan. Methods A total of 166 TB clinical strains were collected to extract the DNA from Jan. 2012 to Mar. 2013 in Tangshan Tuberculosis Hospital. IS6110- PCR fingerprinting method was to be used to identify dominant genotype, and then further identify whether the dominant genotype was "Beijing genotype". Results Beijing genotype was dominated in Tangshan. The genotypes of these isolated strains could be divided into three main types, in which 51 strains belonged to type Ⅰ , 88 strains were type Ⅱ ,11 strains were type Ⅲ and other types were scattered. The dominant genotype was higher in rural than those in urban; was higher in 30 to 60 age group than other age groups. Conclusion Mycobacterium tuberculosis genotype of Tangshan shows aggregation and distribution of "Beijing genotype" dominated. IS6110 - PCR fingerprinting method is efficacious for identifying Mycobacterium tuberculosis genotype in Tangshan.
