胎盘生长因子在宁心利肺汤干预肺动脉高压动物模型中的作用

Effect of Ningxin Lifei Decoction on Plasma Levels of Placenta Growth Factor in Animal Model with Pulmonary Hypertension

摘要 目的建立肺动脉高压大鼠模型,观察宁心利肺汤对大鼠肺动脉压及胎盘生长因子的影响。方法 30只Wistar大鼠10只为正常组,20只行野百合碱注射,建立肺动脉高压的动物模型,治疗组10只给予宁心利肺汤干预,对照组10只灌生理盐水。4周后漂浮导管测肺动脉平均压、右室收缩压,留取静脉血检测胎盘生长因子处死大鼠,计算右室肥大指数。观察肺动脉病理形态学改变。肺血管直径<300μm管壁的厚度,计算管壁相对厚度指数。结果治疗组右室收缩压、肺动脉平均压、肺血管壁相对厚度指数、胎盘生长因子较对照组显著下降,差异有统计学意义(P<0.05)。结论宁心利肺汤有很好地降低肺动脉压,改善肺血管重构的作用,可能与对胎盘生长因子的影响有关。 Objective To establish a rat model with pulmonary hypertension and observe the intervention effects of Ningxin Lifei decoction(NLD) on pulmonary hypertension and expression of placenta growth factor. Methods In 30 wistar rats,10 were in the nor mal group and 20 rats were injected monoerotaline to establish animal model with pulmonary hypertension. The treatment group (n= 10) was given NLD by gastric perfusion. The control group (n= 10) was given physiological saline. Mean pulmonary arterial pressure and right ventricular end systolic were measured via a catheter after 4 weeks. The plasma levels of placenta growth factor were detec ted. The rats were killed and right ventricular hypertrophy index were calculated. The pulmonary vascular remodeling and tuba wall thickness of pulmonary vascular diameter〈300 um were observed. The tube wall thickness index was calculated. Results The pul monary artery mean pressure, left ventricular end systolic and right ventricular hypertrophy index in treatment group vere reduced more than control group(P^0.05). Similarly,so did for MT% and the plasma levels of placenta growth factor. Conclusion NLD was effective to reduce pulmonary arterial pressure and inhibit the pulmonary arterial remodeling. The mechanism might be related with the effect of plaeenta growth factor.
