摘要
目的:研究临床图像引导放射治疗模式下锥形束CT(Cone Beam CT,CBCT)对肿瘤细胞A-549的细胞周期影响,并观察CBCT辐照后采用大剂量照射是否引起肿瘤细胞适应性反应。方法:将体外培养的A-549细胞分为对照组(不照射)、CBCT组(行CBCT照射)、2 Gy组(单纯行2 Gy照射)和CBCT+2 Gy组(CBCT后30min行2 Gy照射),各组照射后经过6h、12h、24h孵育,收集细胞,流式细胞仪检测细胞周期。结果:与对照组相比,CBCT组未发现A-549细胞周期的统计学变化(P>0.05);CBCT+2 Gy组与对照组比较出现了G1期阻滞;CBCT组与2 Gy组比较,未出现A-549肿瘤细胞适应性反应(P>0.05)。结论:CBCT剂量不会引起A-549细胞的周期改变,并且临床治疗模式下CBCT剂量不会引起A-549肿瘤细胞适应性反应,从而不会降低对该肿瘤细胞的杀灭作用。
Objective: To observe the effect of Cone Beam Computer Tomography (CBCT) radiation on the cell-cycle of A-549 cells and to study whether adaptive response ofA549 cells was induced by low-dose radiation.Methods: A-549 cells cultured in vitro, were divided into four groups: control group(no radiation),CBCT group(CBCT radiated),2 Gy group(only 2 Gy dose was radiated) and CBCT+2 Gy group (CBCT and 2 Gy were both conducted,the time internal was 30 min).A-549 cells were collected after 6h,12h and 24h.Then the cell-cycle status was detected using a flow cytometry. Results: Compared with the control group, cell-cycle status of CBCT group did not show statistical differences (P〉0.05).Cells radiated with 2 Gy showed that the percentage of cells in G~/M phase was enlarged. But whether or not take CBCT radiation before 2 Gy radiation did not show statistical differences (P〉0.05).Conclusions: In Image-Guided Radiation therapy, the dose of CBCT did not change the cell-cycle status and induce adaptive response ofA-549 cells, so, it would not weaken the tumor cells extermination.
