甘草酸对人胶质瘤U251细胞和NF-κB的抑制作用 被引量:2

摘要 目的探讨甘草酸(Glycyrrhizic acid,GA)对人胶质瘤细胞的抑制作用及其可能的机制。方法将不同浓度甘草酸作用于U251胶质瘤细胞系,分别用CCK8和平板克隆的方法检测甘草酸对U251增殖及克隆形成能力的影响,并采用Hoechst 33258染色和Annexin-V/PI流式细胞术检测凋亡的变化,免疫印迹及免疫荧光法检测核因子κB(NF-κB)活性亚基p65核内表达的变化。结果经甘草酸处理后,U251细胞生存率降低,克隆形成能力减弱,凋亡细胞增多,并呈现一定的时间、剂量依赖性。不同浓度组甘草酸作用48小时明显降低胶质瘤细胞NF-κB核内表达水平。结论甘草酸能够明显抑制人胶质瘤细胞系U251增殖,并诱导其凋亡,其机制可能与抑制NF-κB活性有关。 Objective To investigate the effect and mechanisms of Glycyrrhizic acid (GA) on human glioma U251 cells. Methods U251 ceils were treated with GA at various concentrations, the effects of GA on cell proliferation and colony formation of U251 cell were measured by CCK8 assay and plate clone-forming test, respectively. And then, cell apoptosis was detected by Hoechst 33258 staining and flow cytometry with Annexin V/PI double staining. The expression of p65 in the nucleus, the active subunit of nuclear factor kappa B(NF-KB) ,was measured by Western blot and immunofluorescence. Results Compared with the control group, U251 cell survival rate and colony formation were decreased in a time- and dose-dependent manner . The apoptotic ratio of GA group was significantly higher than that of the control group. Besides, the expression level of p65 in the nucleuswas reduced after GA treatment. Conclusion GA may inhibit the proliferation and induce apoptosis on human glioma U251 cells via NF-κB mediated mechanism.
