摘要
目的 构建呈现肿瘤相关抗原P815A的噬菌体颗粒 ,在体诱导针对肥大细胞瘤P815的特异性细胞毒性T细胞 (CTL)反应。方法 构建呈现CTL表位P1A35~ 4 3 的噬菌粒载体 ,大量制备呈现此表位的重组噬菌体颗粒 ,小剂量、无佐剂皮下免疫DBA/2小鼠 (H 2 d) ,采用51Cr释放法、ELISPOT观察针对肥大细胞瘤P815的CTL反应。结果 重组噬菌体呈现颗粒疫苗在体诱导了针对弱抗原P815A的CTL反应。结论 丝状噬菌体呈现疫苗增强肿瘤相关抗原的免疫原性 。
Objective Genetically engineer hybrid filamentous phage particles containing mutant gVⅢ with the P815A epitope and induce of the mouse mastrocytoma P815 specific cytotoxic T Iymphocytes response in vivo . Methods We constructed the hybrid filamentous phage particles, which displayed the P1A 35~43 epitope, and then injected of low doses of the hybrid virus particles into DBA/2 (H 2 d) mice without adjuvants. The anti tumor cytotoxic T Iymphocytes response was investigated by 51 Cr release assay and ELISPOT. Results The peptide epitope in the hybrid bacteriophages primed the tumor associate antigen PLA specific CTL response. Conclusion These data suggest that the filamentous phage display vaccine could be a kind of potentially efficient form to develop the anti cancer vaccine.
