哮喘大鼠淋巴液和血液中Th17细胞百分率升高及IL-23、IL-17的表达增加 被引量:9

An increased of Th17 cells and IL-23 and IL-17 in the lymph and blood of rats with bronchial asthma

摘要 目的验证免疫反应能否改变哮喘大鼠外周血和淋巴液中CD4+IL-17+Th17细胞数量及其相关细胞因子IL-23、IL-17的表达。方法流式细胞仪检测大鼠血液和淋巴液中CD4+IL-17+Th17细胞百分率,ELISA检测血液和淋巴液上清中IL-17、IL-23水平,实时荧光定量PCR(qRT-PCR)检测淋巴液和血液中IL-23 p19 mRNA表达。结果末次激发后各时间点(0、24、48h)收集单个核细胞(MNC),体外培养18 h,探讨哮喘模型中细胞因子产生和CD4+IL-17+Th17细胞变化的影响。哮喘组淋巴液和血液MNC的体外培养体系中CD4+IL-17+Th17细胞百分率和IL-23 p19 mRNA的表达水平均显著高于正常对照组(P<0.05);另外,各组淋巴液MNC的CD4+IL-17+Th17和IL-23 p19 mRNA的表达水平均显著高于血液(P<0.05)。各组淋巴液上清IL-23、IL-17水平均显著高于血液(P<0.05),与正常组比较,哮喘组的细胞因子和体外培养体系中细胞数量明显增加。结论哮喘促使Th17细胞的增殖和释放IL-23、IL-17,可能与其在血液和淋巴液中发挥不同的免疫作用有关。 Objective To investigate whether immune response could change the number of CD4+ IL-17+ Th17 cells andthe concentrations of IL-17 and IL-23 in the serum and lymph of asthmatic rats. Methods Both serum and lymph CD4+ IL-17 + Th17 cell numbers were measured by flow cytometry. The concentrations of IL-17 and IL-23 in the serum and lymph wereassayed by ELISA. The expression of IL-23 pl9 mRNA was examined by quantitative real-time PCR. Results Mononuclearcells (MNC) were collected from the blood and lymph of rats 0,24 and 48 hours after final ovalbumin stimulation and weresubsequently cultured for 18 hours. The serum and lymph of the asthma group produced higher amounts of CD4 +IL-17+ Th17cells, and higher levels of IL-23pl9 mRNA, than those of the healthy controls (P〈0.05). In addition, compared to lymph,serum had significantly lower amounts of CD4+ IL-17+ Th17 cells, decreased expression of IL-23pl9 mRNA, and reduced IL-23 and IL-17 concentratons ( P 〈 0. 05). Cytokines and cell numbers of Th17 cells from the asthma group significantlyincreased compared with that of control group. Conclusion These data suggested that asthma-driven CD4+ IL-17+ Th17-cellproliferation and release of IL-17 and IL-23 may be associated with different immune activities between the serum and lymph.
