摘要
本试验选择犬为试验动物,构建角膜高危病例后进行穿透性角膜移植术(PKP)。术后将45只犬随机分为3组,分别使用环孢霉素、雷帕霉素滴眼液和生理盐水进行滴眼治疗,并分别于术后10d、30d和60d三个时间点每组随机抽取3只术犬,摘眼后进行角膜组织切片,对角膜各层组织结构的形态进行观察。结果显示,生理盐水组术犬角膜随着时间推移,出现了结构层次的变化,细胞破裂、丢失,大量新生血管浸润,内皮细胞丢失。环孢霉素组术犬角膜组织结构无明显改变,基质层出现少量红细胞,内皮细胞轻微丢失。雷帕霉素组术犬角膜组织结构无明显改变,未见新生血管浸润。
dogs were taken as experimental animals in this test and high risk cornea pathologic model were established and PKP were conducted. The forty-five dogs were randomly divided into three groups, fifteen dogs were treated by normal saline, fifteen dogs were treated by CSA and another fifteen dogs were treated by RAPA. Three dogs in each group were randomly selected for corneal histological examine on 10th,30th,60th respectively and the organizational structures of cornea were observed. The result showed that the cornea of normal saline group became worse over time cells were broken down and lost,greater a- mounts of CNVs infiltrated into the cornea, endothelial cells were lost. In the CSA group the corneal was normal except there were few RBCs in stromal layer and a-slight loss of the endothelium. The structure re- mained normal and no CNVs were found in RAPA group.
