摘要 经硫酸铵盐析沉淀并经 DEAE_(32)—纤维素枉色谱分离粗制 IgM,再经聚丙烯酰胺凝胶电泳(PAGE)进一步分离纯化鸡血清 IgM。然后用木瓜蛋白酶水解IgM,经 DEAE_(32)—纤维素柱色谱纯化 IgMFc 重链,再用 IgMFc 重链免疫家兔制取兔抗鸡 IgMFc 重链抗血清。 Rough immunoglobulin M (IgM) in chicken serum was obtained by salting-out withammonium sulfate and DEAT_(32)-cellulose column chromatohgraphy,The IgM was furtherisolated and purified by polyacrylamide gel electrophoresis (PAGE).Then papain was usedto hydrolize the pure IgM and the IgMFc heavy chains were purified by DEST_(32)—cellulosecolumn chromatography.The rabbit anti-chicken IgMFc heavy chain antisra wer preparedafter the rabbits were injected with the IgMFc heavy chain.
