摘要
目的研究两种粉碎方式下所制备腰痛宁粉(马钱子、苍术、麻黄、土鳖虫、川牛膝、僵蚕、甘草、乳香、没药、全蝎)对小鼠急性毒性及耐受性的反应。方法采用扫描电镜、激光粒度仪分析两种腰痛宁粉的微观形貌特征与粒径;以昆明种小鼠为受试对象,采用Bliss法测定两种腰痛宁粉的口服急毒LD50;对小鼠连续给药10 d后,于第11天给小鼠口服急毒LD50剂量的腰痛宁粉并继续观察7 d,记录体质量。采用显著性t检验分析小鼠17 d间体质量差异。结果马钱子单独粉碎与马钱子混合粉碎方式下的腰痛宁粉小鼠口服的LD50分别为314.00 mg/kg与229.68 mg/kg;前者的平均粒径大于后者。结论马钱子单独粉碎方式下的腰痛宁粉安全性略优。
AIM To research the acute toxicity and tolerance response of mice to Yaotongning Powder(Strychni Semen,Atractylodis Rhizoma,Ephedrae Herba,Eupolyphaga seu Steleophaga,Cyathulae Radix,Bombyx Batryticatus,Glycyrrhizae Radix et Rhizom,Olibanum,Myrrha,Scorpio) prepared by two crushing modes.METHODS Scanning electron microscopy and laser particle size analyser were applied to characterizing the microscopic morphology and particle sizes of the two kinds of Yaotongning Powder.Bliss method was used to test oral LD50 of Kunming mice by gavage for ten days.The mice were given the two kinds of Yaotongning Powder of LD50 dose on the eleventh day and were consecutively observed for seven days.Significant t-test was applied to analyzing the change of the mice's weights during the 17 days.RESULTS The average particle size of Yaotongning Powder prepared by single grinding Strychni Semen was greater than that of Yaotongning Powder prepared by mix grinding Strychni Semen with other herbs.Their LD50 were 314.00 mg/kg and 229.68 mg/kg,respectively.All the mice behaved normally and no deaths occurred during the 17 days.CONCLUSION The safety of Yaotongning Power produced by single grinding Strychni Semen is slightly better than that by mix grinding Strychni Semen with other herbs.
