摘要
目的研究蕨麻乙醇提取物对缺血再灌注损伤心肌iNOS、eNOS蛋白和NO水平的影响,探讨其作用机制。方法建立垂体后叶素所致急性心肌缺血再灌注模型,采用比色法检测血清及心肌组织NO含量,采用免疫组织化学方法检测心肌组织iNOS蛋白水平,Westernblot方法检测心肌组织eNOS蛋白水平。结果蕨麻乙醇提取物可抑制iNOS蛋白的表达,但可升高eNOS蛋白的表达,提高血清及心肌组织中NO水平。结论蕨麻乙醇提取物可能通过抑制iNOS活性,升高eNOS活性,促进有益的NO生成,从而减轻心肌缺血再灌注损伤。
Objective To explore the effects of the Potentilla anserine L. alcohol extract on the content of NO and the protein levels of iNOS and eNOS in myocardial ischemia-reperfusion injury mice. Methods The myocardial ischemia-reperfusion injury model was developed; the content of NO was detected by spectrophotometry, the protein levels of iNOS and eNOS were determined respectively by immunohistochemistry and Western blot. Results The content of NO in cardiac muscles and serum increased significantly, The protein level of iNOS in cardiac muscle was down-regulated significantly by Potentilla anserina L. while the level of eNOS in cardiac muscles was up-regulated obviously. Conclusion The Potentilla anserine L. alcohol extract is considered being effective in protecting the myocardium against ischemic-reperfusion injury by inhibiting activity of iNOS and increasing activity of eNOS, as well as promoting of beneficial NO production.
