摘要
为评估糖及乙醇对种子液活力的影响,制备含不同糖源无乙醇和添加乙醇的种子液,将种子液接种至生产培养基中,通过测定纤维素产量来评价其活力;同时将这种影响分为静置和振荡培养2种模式下进行研究。结果显示:种子液代数增加,无论是静置方式还是振摇方式,种子液活力随转接次数增加而下降,但这种下降的趋势因糖的种类不同而异,以2%葡萄糖为糖源时,下降趋势更缓,如果添加1%vol的乙醇,因为乙醇有增菌作用,下降趋势得到缓解。静置方式与振摇方式相比,更能保持菌种的活力。
In order to evaluate effects of sugars and ethanol on viability of Gluconacetobacter xylinus in stock culture, stock culture with different sug- ars, namely sucrose, glucose was prepared. Ethanol was added if needed, and it was stationarily cultivated or shaking, then inoculated into liquid medium. Based on production of bacterial cellulose in liquid medium, viability of stock culture was evaluated. The results showed that viability of stock culture had a downward trend during successive subculture, which varied with types of sugars. When 2% glucose was used, there would be the least downward trend, and when 1% ethanol was added into stock culture with 2% glucose, downward trend could be relieved because of benefits of ethanol on propagation of Ga, xylinus. Compared the viabilities of stock cultures in stationary cultivation with shaking, it was found that the former was better.
