摘要
目的 探讨肿瘤细胞上的主要组织相容性复合体 (majorhistocompatibilitycomplex ,MHC)分子在诱导自体口腔鳞癌细胞毒T淋巴细胞 (cytotoxicTlymphocyte ,CTL)中的作用。方法 对 9例口腔鳞癌中的肿瘤细胞表面人类白细胞抗原 (humanleukocyteantigen ,HLA) ABC和HLA DR抗原进行检测 ,将其中 5例HLA ABC抗原表达 >5 0 %和HLA DR抗原表达 >35 %的病例作为研究对象 :①自体肿瘤细胞经处理后作刺激细胞 ,并用MHC单抗阻断刺激细胞上的HLA ABC和 (或 )HLA DR抗原 ,检测自体肿瘤细胞诱导的CTL的特异性杀伤活性及肿瘤坏死因子 (tumornecrosisfactor,TNF) α的分泌水平。②阻断靶细胞上HLA ABC和 (或 )HLA DR抗原 ,观察MHC单抗对自体刺激细胞 (未加单抗处理 )诱导的CTL细胞毒的抑制作用。结果 ①各病例的各实验组刺激细胞诱导的CTL杀伤活性均显著低于对照组 ,依次为 :DR抗原阳性刺激细胞组 >ABC抗原阳性刺激细胞组 >ABC和DR抗原均阴性刺激细胞组 ,抗ABC单抗可完全阻断CTL特异性分泌TNF α ;②各实验组单抗对CTL的杀伤活性的平均抑制率分别为 :抗ABC单抗为 5 2 % ;抗DR单抗为 2 7% ;抗ABC单抗 +抗DR单抗为 72 %。 结论口腔鳞癌细胞上的MHC I。
Objective To investigate the role of major histocompatibility complex(MHC) molecules in the induction of cytotoxic T lymphocyte(CTL) from oral squamous cell carcinoma. Methods ①The treated autologous tumor cells served as stimulating cells,on which the MHC mAb was used to block the HLA ABC and/or HLA DR antigens.The specific cytotoxicity of CTL induced by stimulating cells and the level of released TNF α were then detected. ② By blocking the HLA ABC and/or HLA DR antigens on target cells , the inhibition effect of MHC mAb on CTL cytotoxicity induced by autologous stimulating cells (not treated with mAb) was measured. Results ①The cytotoxicity induced by stimulating cells in experimental groups was significantly lower than that of the control group. The successive order was: the DR antigen positive stimulating cell group > ABC antigen positive stimulating cell gooup > both ABC and DR antigen negative stimulating cell group . The specific TNF α released by CTL was completely blocked by anti ABC mAb.②The average inhibition rate by mAb on cytotoxicity of CTL was: 52%,27% and 72% by anti ABC mAb, anti DR mAb, and anti ABC mAb plus anti DR mAb respectively. Conclusion Both MHC class I and class Ⅱantigens on OSCC cells play an important role in inducing CTL.
