补中益气丸对急性运动疲劳大鼠脑组织线粒体呼吸链复合体及ATP酶活性的影响 被引量:8

Effect of Buzhongyiqi Pill on the activities of respiratory chain complexes and ATPase in the brain mitochondria of acute exercise-induced fatigue rats

摘要 目的:观察补中益气丸对急性运动疲劳大鼠脑组织线粒体呼吸链复合体及ATP酶活性的影响。方法:将24只SD大鼠进行随机分组,分为安静对照组、急性运动组和补中益气丸组,每天按3g/kg的剂量灌胃补中益气丸混合液一次,药物补充一周后进行实验。急性运动为大强度间歇性跑台运动,速度为27m/min,坡度为0,总运动时间90min,每运动10min间歇1min。采用双抗体夹心法酶联免疫分析(ELISA)测定大鼠脑组织中线粒体呼吸链复合物活性浓度,采用无机磷法测定大鼠脑组织中Na+-K+-ATPase和Ca2+-ATPase活性浓度。结果:大鼠脑组织呼吸链复合体Ⅰ和Ⅲ各组之间均无显著性变化。与安静对照组比较,急性运动组呼吸链复合体Ⅱ活性、Na+-K+-ATPase和Ca2+-ATPase活性均显著降低;与急性运动组比较,补中益气丸组(3g/kg)呼吸链复合体Ⅱ、呼吸链复合体Ⅳ、Na+-K+-ATPase和Ca2+-ATPase活性均显著提高。结论:补中益气丸能改善急性运动后大鼠脑组织呼吸链复合体Ⅱ、Ⅳ及ATP酶活性。 Objective: To observe the effect of Buzhongyiqi Pill on the activities of respiratory chain complexes and ATP enzyme in the brain mitochondria of acute exercise-induced fatigue rats.Methods: 24 SD rats were randomly divided into two groups,divided into control group,acute exercise group and Chinese medicine group.Chinese medicine group according to 3g per kilogram of body weight per day dose adminis tered supplementary Buzhongyiqi Pills mixture(0.3g / ml) once,pre-exercise supplement for a week.Acute exercise for high intensity intermittent exercise,speed of 27m / min,the slope is zero,total exercise time 90min,10 min interval of 1 min per movement.Activity of mitochondrial respiratory chain complexes concentration in rat brain tissue was measured by using double antibody clip art enzyme-linked immunoassay(ELISA) determination.Na+-K+-ATPase and Ca2 +-ATPase activity concentrations were measured in rat brain tissues by inorganic phosphorus method.Results: There were no significant changes of rat brain respiratory chain complex I and III in above three groups.The activity of respiratory chain complex Ⅱ and ATPase of acute exercise group was significantly lower than that of control group;the activity of respiratory chain complex Ⅱ,IV and ATPase of Chinese medicine group(3g / kg) was significantly higher than that of acute exercise group.Conclusion: The supplement of the Buzhongyiqi Pills could improve the activity of rat brain respiratory chain complex II,IV and ATPase after acute exercise.
