不同添加剂对自制尿液室内质控物质量的影响

摘要 目的为自制尿液复合型室内质控物选择最佳添加剂。方法分析一种包含尿N-乙酰-B-D-氨基葡萄糖苷酶(NAG)和尿微量清蛋白(UMA)两个项目的自制复合室内质控物的酶稳定剂、蛋白稳定剂及叠氮钠等添加剂对其稳定性及均一性的影响。结果在-20℃保存温度下,尿NAG项目的稳定天数:无添加,72d;叠氮钠,169d;酶稳定剂,大于365d。同样条件下,尿UMA项目的稳定天数:无添加,99d;叠氮钠,207d;蛋白稳剂定,大于365d。质控物各个保存阶段稳定的同时测得的瓶间均一性良好。结论在制备尿NAG和UMA两项复合室内质控物的同时添加酶稳定剂、蛋白稳定剂和叠氮钠能延长质控物的稳定时间,提高质控物的质量。 Objective To choose the best additive for self-made internal QC mixture for urine detection. Methods Clinical fresh urine samples of kidney patients were collected and made into QC multiplex material for NAG and UMA measurments, then the homogeneity and stability were tested for quality control materials with different additives, which were enzyme stabilizer, protein stabilizer and sodium azide respectively. Results The number of stable days for NAG were 72 d(no additives), 169 d(sodium azide) ,〉365 d(enzyme stabilizer),respectively. The number of stable days for UMA were 99(no additives),207(sodium azide),〉 365(protein stabilizer). The homogeneity among samples was good. Conclusion Enzyme stabilizer, protein stabilizer and sodium azide can extend the stable days of self-made internal QC material for NAG and UMA measurements.
