钛合金微粒负荷成骨细胞对肿瘤坏死因子-α、白介素-1β和白介素-6的影响 被引量:2

摘要 目的探讨不同浓度钛合金微粒对成骨细胞肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)和白介素-6(IL-6)的影响,了解骨溶解后炎性反应因子和成骨细胞间的关系。方法将培养成骨细胞分为空白对照组和不同浓度钛合金微粒处理组:5 mg/mL(A组)、10 mg/mL(B组)、15 mg/mL(C组)。采用Elisa法检测各组细胞培养72 h后TNF-α、IL-1β和IL-6含量的变化。结果培养72 h后,与对照组相比,各实验组TNF-α、IL-1β和IL-6含量均增加,差异有统计学意义(均P<0.05)。结论钛合金微粒可增加成骨细胞TNF-α、IL-1β和IL-6含量,刺激破骨细胞导致骨溶解可能是关节松动的机制之一。 Objective Tissues of around prosthesis contains a lot of inflammatory cytokines in aseptic loosening, osteoblasts are mediated role unclear. This paper investigate different concentrations of Titanium alloy particles (TAPs) on osteoblasts TNF-α, IL-1β and IL-6, and further understanding of osteolysis and inflammatory factor between osteoblasts. Methods Osteoblasts were cultured in medium containing different concentrations of TAPs. Experiments were cultured in medium with TAPs, while osteoblasts in groups A, B and C were cultured in medium containing TAPs at 5 mg/ml, 10 mg/ml and 15 mg/ml, respectively. Elisa was used to assay effects of inflammatory cytokines after 72 h incubation with TAPs. Results Compared with the control cells, after 72 h-incubation witll TAPs, the TNF-α, IL-1β and IL-6 of osteoblasts in group A, B or C was significantly increased with comparison of the control cells in control group (P 〈 0. 05). Conclusion TAPs increased level of TNF-α, IL-113 and IL-6 on osteoblasts, and these inflammatory cytokines stimulated bone resorption by osteoclast. We hypothesized that inflammatory cytokines regulate osteoblast result in aseptic loosening may be one of the mechanisms.
