摘要
目的 观察纯化的蜂毒肽对兔免疫性关节炎的病理过程是否有抑制作用。方法 采用柱层析方法纯化粗制蜂毒 ,以除去具有溶血及化学刺激作用的杂质。纯化物经CD光谱法和荧光光谱法 ,高压液相色谱分析及氨基酸序列分析。造成家兔卵蛋白性关节炎模型 ,给予蜂毒肽肌肉注射 ,观察组织炎症反应的变化。并测定兔血红蛋白水平 ,进行了体外溶血实验。结果 高压液相色谱分析显示单峰 ,CD光谱法和荧光光谱法、高压液相色谱分析氨基酸序列分析显示纯化物氨基酸序列与文献报道峰毒肽一致。实验组应用纯化蜂毒肽后 ,滑膜炎症明显减轻。溶血实验显示 ,纯化蜂毒肽只有较弱的溶血作用。结论 纯化蜂毒肽对兔免疫性关节炎具有良好的抗炎作用 ,并且溶血作用较弱。
Objective To investigate whether the purified melittin has anti-inflammation effect on immuno-arthritis of rabbits.Methods Natural rough melittin was purified by column chromatography to eliminate the hemolytic abilities.Results The products were analyzde by fluorescense and circular dichroism spectrum and anti- inflammation effect of it on immune arthritis of rabbits were tested.Conclusion The results showed that melittin possesses significant anti-inflammation effect on immune arthritis of rabbits and the hemolytic activity is greatly reduced.
