摘要 目的 探讨硝酸纤维素膜为固相载体的酶免疫测定法检测甲胎蛋白 (AFP)。方法 以硝酸纤维素膜代替常规ELISA中的固相载体聚苯乙烯板 ,其余步骤按ELISA方法操作。结果 以AFP标准品作试验 ,进行方法学探索 ,测出最佳包被抗体浓度为 10 0mg·L-1,酶标抗体稀释度为 1∶40 0 0。不同来源不同孔径的硝酸纤维素膜无差异 ;同时发现包被过程中的封闭处理能减少非特异性反应 ;保存试验证明包被抗体的硝酸纤维素膜至少可保存 2周。结论 该法具有特异性强、重复性好、灵敏度高的特点 ,通过与常规ELISA检测血清标本AFP含量值的比较 ,表明该法可半定量测定AFP的含量 ,尤其适用于标本的初步筛选。 Objective The enzyme immunoassay in which the solid state carrier is served as the nitrocellulose membrane was used to detect α-fetoprotein (AFP). Methods The solid state carrier with nitrocellulose membrane substituting for polystyrene plate among the convention ELISA, the other step was operated according to ELISAs method. Results The optimum bundle by antibody consistency was 100 mg·L -1, and enzyme linked antibody of dilution 1∶ 4 000. The experiment indicates that the nitrocellulose membrane of different apertures and different sources did not have clear difference. The quench could reduce the nonspecific reaction by wrapping. Preserving experiment proved that the nitrocellulose membrane wrapped by antibody could be stored at least two weeks.Conclusion Experiment proves that this method possesses good specialties, repetitions and high sensitivity. Compared with convention ELISAs checkout serum specimen AFPs content value, it is suggested that this method can measure the content of AFP quantitively, particularly suitable for the specimen screen.
