摘要
测定了两种不同生长环境的齐口裂腹鱼和草鱼肌肉组织的乳酸脱氢酶和苹果酸脱氢酶活力,采用聚丙烯酰胺凝胶电泳分析了这两种鱼类的心脏、肝胰脏、鳃、脑及肌肉5种组织的乳酸脱氢酶同工酶谱。结果显示:齐口裂腹鱼的苹果酸脱氢酶/乳酸脱氢酶活力显著高于草鱼(P<0.05),齐口裂腹鱼和草鱼乳酸脱氢酶同工酶谱具有明显的物种特异性和组织特异性,草鱼各组织中乳酸脱氢酶的迁移率明显快于在齐口裂腹鱼各组织中,两种鱼的心脏和脑中乳酸脱氢酶的含量比较丰富;两种鱼的心、脑组织中乳酸脱氢酶1-乳酸脱氢酶3占优势,乳酸脱氢酶4和乳酸脱氢酶5在肌肉组织中占优势。
In this study, lactate dehydrogenase(LDH) and malate dehydrogenase(MDH) activities were evaluated in muscle from two cyprinid fish, Schizothorax prenanti and grass carp Ctenopharyngodon idellus inhabiting under different environments. The isozyme patterns of LDH were also determined in heart, hepatopancreas, gill, brain and muscle in S. prenanti and grass carp by SDS-PAGE. The results showed that there were significantly higher activities of MDH and LDH in S. prenanti than those in grass carp(P〈0.05). Isozyme pattern analysis revealed that expression of LDH was species- and tissue-specific. The grass carp had significantly more rapid migration rate of LDH in all tissues than S. prenanti did. Both fish showed higher LDH contents in heart and brain than in other tissues. LDH1, LDH2 and LDH3 were found to be prominent in hearts and brains in both fish, while LDH4 and LDH5 prominent in muscle.
