多囊卵巢综合征患者血清核因子-κB和转化生长因子β1的表达及其与胰岛素抵抗的相关性研究 被引量:16

Serum Nuclear Factor-κB and Transforming Growth Factor-a1 and Those Relationship with Insulin Resistance in Polycystic Ovary Syndrome

摘要 目的:探讨多囊卵巢综合征(PCOS)患者血清中核因子-κB(NF-κB)、转化生长因子β1(TGF-β1)的表达,及NF-κB、TGF-β1与胰岛素抵抗(IR)的相关性。方法:收集108例PCOS患者作为PCOS组,47例正常女性作为对照组,根据体质量指数(BMI)将其分为肥胖组(BMI≥28 kg/m2)与非肥胖组(BMI<28 kg/m2),根据胰岛素抵抗指数(HOMA-IR)将PCOS患者分为IR组(HOMA-IR≥2.69)及非IR组(HOMA-IR<2.69);应用酶联免疫法测量受试者血清中NF-κB、TGF-β1水平,同时对入组患者进行葡萄糖耐量(OGTT)及胰岛素释放(IRT)测定。结果:①PCOS患者血清中NF-κB、TGF-β1明显高于对照组(P<0.05),PCOS-IR组NF-κB、TGF-β1明显高于PCOS非IR组(P<0.05);②Pearson相关分析显示,NF-κB、TGF-β1与PCOS患者的雄激素(T)水平、BMI、HOMA-IR呈正相关(P<0.05)。结论:NF-κB、TGF-β1与PCOS患者T水平、IR密切相关,TGF-β1与NF-κB亦呈正相关,NF-κB、TGF-β1与PCOS密切相关。 Objective: To study the expression of serum nuclear factor-kB (NF-kB) and transforming growth factor-f3 I(TGF-β1) in polycystic ovary syndrome (PCOS) patient and analyze the correlation among insulin resistance (IR), NF-kB and TGF-β 1. Methods: A total of 108 female patients with PCOS were recruited as PCOS group and compared with 47 healthy women from control group. Participants were divided into two groups based on body mass index (BMI), BMI ≥ 28 kg/m2 was considered as the obese group, BMI〈28 kg/m2 was considered as the non-obese group; PCOS women were divided into two groups using the criterion of HOMA-IR≥ 2.69 as IR group and HOMA-IR〈2.69 as non-IR group. Serum NF-kB and TGF-β1 levels were measured using euzymelinked immunosorbent assay (ELISA). Oral glucose tolerance test (OGTT) and insulin sensitivity were also assessed in the selected people. Results: 1) The PCOS group presented significantly higher serum NF-kB and TGF-β1 than those in the control (P〈0.05), while the two factors are obviously higher in PCOS-IR group than in PCOS-non-IR group (P〈0.05). 2) Pearson analysis revealed that NF-kB and TGF-β1 of the PCOS group had a positive correlation with HOMA-IR, BMI and testosterone (T) level (P〈0.05). Conclusion: NF- kB is also positively correlated with TGF-β1, while they have a strong link with IR and T level in women with PCOS.
