摘要
目的了解TLR4基因Asp299Gly多态性与消化系统肿瘤关系。方法检索中文及英文数据库中有关TLR4基因Asp299Gly多态性与消化系统肿瘤关系的研究文献,根据异质性分析结果选择固定效应或随机效应模型计算合并OR值,同时进行敏感性分析及发表偏倚分析。计算采用Stata 10.0统计软件。结果共10篇文献12项研究符合条件,剔除对照组不符合H-W平衡的研究后,AG+GG/AA基因型与消化系统肿瘤关系的合并OR值为1.29,95%CI为0.89~1.86(P=0.18);G/A等位基因与消化系统肿瘤关系的合并OR值为1.44,95%CI为0.92~2.27(P=0.11);AG+GG/AA基因型与胃癌关系的合并OR值为1.77,95%CI为1.34~2.34,有统计学意义(P<0.05);G/A等位基因与胃癌关系的合并OR值为1.76,95%CI为1.35~2.30,有统计学意义(P<0.05)。结论TLR4基因Asp299Gly多态性的G等位基因增加胃癌的患病风险,但对其他消化系统肿瘤可能无影响。
Objective To understand the association between Asp299Gly polymorphism in TLR4 gene and digestive sys- tem tumors. Methods Literatures concerning the aforementioned topic were collected from database in Chinese and English. The pooled ORs were calculated in the reasonable model (fixed effect model or random effect model) according to heterogeneity analysis by Stata version 10.0. Sensitivity analysis and publication basis were performed. Results Twelve case-control studies in ten literatures met the criteria. After excluding an article that deviated from Hardy-Weinberg equilibrium in controls, the meta- analysis results showed the pooled ORs of genotype AG+GG/AA and allele G/A of case vs. control on Asp299Gly and digestive system tumor were 1.29 (95%CI:0.89-1.86) (P=0.18),1.44 (95%CI:0.92-2.27) (P=0.11), respectively. And the pooled ORs of genotype AG+GG/AA and allele G/A of case vs. control on Asp299Gly and gastric cancer were 1.77(95%CI:1.34--2.34) (P〈0.05), 1.76(95% CI:1.35--2.30) (P〈0.05), respectively. Conclusion The G allele of TLR4 gene Asp299Gly polymor- phism may increase the risk of gastric cancer, but may not associate with other-type tumors of digestive system.
