钛颗粒诱导的兔人工关节假体松动模型制备 被引量:1

ESTABLISHMENT OF PROSTHESIS LOOSENING MODEL INDUCED BY TITANIUM PARTICLE IN RABBIT

摘要 目的制备一种由较大直径钛颗粒介导的生物性人工关节假体松动动物模型,初步验证该模型在相关基础及临床研究中的应用价值。方法将10只平均体质量为3.0kg的新西兰大白兔随机分为对照组和实验组,两组动物均从双侧的股骨膝关节面将钛棒假体沿轴线方向植入到髓腔内,实验组在假体植入前于其表面均匀涂抹50μg直径20μm的钛颗粒。术后12周处死动物,处死前2d行X线检查,观察假体周围成骨及骨溶解程度。动物处死后取包含假体的股骨段及膝关节滑膜囊,股骨段制作成厚度为50μm的骨磨片,行Masson Trichrom Goldner染色及Kossa染色;关节滑膜做石蜡切片,行苏木精-伊红染色,以观察骨组织形态学变化及炎性反应情况。结果两组动物X线检查均未见明显骨溶解。组织形态学观察显示,实验组的骨假体接触率、假体周围骨体积分数、骨皮质处钙盐沉积岛平均面积均低于对照组,而钙盐沉积岛数量大于对照组,差异有统计学意义(t=4.083~10.200,P<0.05),且实验组假体周围及关节滑膜的炎性反应较对照组明显活跃。结论成功制备了较大直径钛颗粒介导的新型人工膝关节假体松动模型,经验证该模型制作简便,成本低廉,易于推广。 Objective To create an artificial joint loosening model induced by large diameter titanium debris in rats and assess its value in related basic and clinical research. Methods Ten New Zealand white rabbits (average weight of 3.0 kg) were equally randomized to control and experimental group. The bilateral distal femurs of each rabbit were implanted a titanium rod into medullary cavity. In the experimental group, each rod was evenly smeared with 50 μg titanium debris before implanted. Two days before the kill of the rabbits, X-ray films were taken, and the ossification and osteolysis around the rod observed. All the animals were killed after 12 weeks, the femurs and synovium of the knees were collected. The femurs including the titanium rod were made into 50 μm ground bone slices and stained by Masson Trichrom Goldner staining and Kossa staining to observe the index of bone histomorphology and inflammatory reaction. Results No obvious osteolysis was noted in both groups. Compared with the con- trol group, the bone-implant contact rate, the volume percentage of bone around implant and the average area of calcium deposition were less in the experimental group, the numbers of calcium deposition were more (t = 4.083-10.200, P〈0.05). The inflammato- ry reaction of in experimental group was more active than the control group. Conclusion A new type animal model of artificial joint loosening induced by large diameter titanium is created. It is recognized that the model has advantages of being easy to build, easy to popularize and low cost.
