阿托伐他汀对大鼠脑出血后血肿周围血管内皮生长因子表达的影响 被引量:4

摘要 目的研究阿托伐他汀对大鼠脑出血后血肿周围脑组织血管内皮生长因子(VEGF)表达的影响。方法 SD大鼠108只,随机分成正常对照组、模型组和阿托伐他汀治疗组,每组各36只。模型组和阿托伐他汀治疗组大鼠采用改良2次注血法,将自体血注入到脑左侧基底节区,诱导大鼠脑出血模型;建模成功后各组大鼠分别给予2 ml生理盐水或阿托伐他汀(20 mg/kg)灌胃,共7 d。分别于出血后6 h、12 h、1 d、3 d、5 d和7 d对各组大鼠行神经功能缺损评分,免疫组织化学染色检测各组大鼠血肿周围脑组织VEGF的表达,脑出血后第3和7天分别测定脑组织含水量。结果建模后6~12 h,模型组及阿托伐他汀治疗组大鼠的神经功能评分明显升高,其后评分逐渐下降;在建模后第5和7天,阿托伐他汀治疗组神经功能评分较模型组明显降低(P<0.05)。建模后6 h,血肿周围脑组织表达VEGF的阳性细胞即增多;与模型组比较,阿托伐他汀治疗组大鼠建模后6 h、12h、1 d、3 d、5 d和7 d血肿周围组织表达VEGF的阳性细胞均明显升高(P<0.01)。与正常对照组相比较,模型组及阿托伐他汀治疗组大鼠建模后第3天脑组织含水量明显增高(P<0.05);与模型组相比,阿托伐他汀治疗组大鼠建模后第7天脑组织含水量明显降低(P<0.05)。结论阿托伐他汀能通过促进血肿周围组织VEGF表达,促进大鼠脑出血后神经功能的恢复。 Objective To study the effect of atorvastatin on the expression of vascular endothelial growth factor(VEGF) on perihematoma brain tissue after cerebral hemorrhage in rats.Methods A total of 108 SD rats were randomly divided into 3 groups: normal control group,model group,and atorvastatin treatment group.Rats in the model group and atorvastatin treatment group received intracranial injection of autologous arterial blood 50 μl into the left basal ganglia by two-step injection method.Rats in normal control group and model group were fed 2 ml saline per day for 7 days,and rats in atorvastatin treatment group were administered 2 ml saline containing atorvastatin(20 mg / kg) per day for 7 days.The neurological deficits were assessed according to the modified neurological severity scores,then the expression of VEGF in the area surrounding the hematoma in the left basal ganglia were examined at 6 h,12 h,1 d,3 d,5 d and 7 d respectively by immunohistochemistry.The contents of brain water were measured at 3 d and 7 d,respectively in three groups.Results Modeling after 6-12 h,the neurological function score of rats in model group and atorvastatin treatment group increased significantly,and then gradually declined.At 5 d and 7 d after modeling,the neurological function score of rats in atorvastatin treatment group was significantly lower than that in the model group(P 0.05).The expression of VEGF positive cells was found to increase surrounding the area of hematoma after 6 h hemorrhage.The expression of VEGF cells in surrounding area of hematoma increased significantly in atorvastatin treatment group compared with that in model group at each examination point(P 0.01).Compared with normal control group,the contents of brain water at 3 d after modeling increased significantly in model and atorvastatin treatment groups(P 0.05),and those in atorvastatin treatment group reduced significantly compared with model group at 7 d after modeling(P 0.05).Conclusion Atorvastatin can promote recovery of neural function after cerebral hemorrhage in rats by up-regulating the expression of VEGF in brain tissue arround hematoma.
