摘要
目的探讨转化生长因子β1(TGF-β1)及其受体TβRⅠ、TβRⅡ与矽肺发生、发展的关系。方法 无特定病原体级健康成年Wistar大鼠70只,随机分成2组,每组各35只,实验组采用非暴露式经气管染尘法制备矽肺动物模型,对照组注入等体积的生理氯化钠溶液,分别于染尘后第1、3、7、14、28天各处死7只,应用免疫组化法结合图像分析处理系统检测TGF-β1、TβRⅠ、TβRⅡ在矽尘致大鼠肺纤维化中的表达。结果 TGF-β1、TβRⅠ和TβRⅡ广泛分布于肺组织内。实验组肺组织TGF-β1表达量高于对照组,差异有统计学意义(P<0.01),表达量在第7天达到高峰;与对照组比较,实验组肺组织TβRⅠ和TβRⅡ表达量在染尘后第14、28天增高,差异均有统计学意义(P<0.01)。结论 TGFβ1、TβRⅠ和TβRⅡ在矽尘所致的肺纤维化中具有重要的作用。
Objective To study the expression level and significance of TGF-β and its receptor I ( Tβ3R I ) and receptor II (TβR I1 ) in silicosis. Methods Seventy healthy spectific pathogen free Wistar rats were randomly divided into control group and model group, with 35 rats in each group. Silicotic animal models were established by intratracheal dust injection and the control group was injected with normal saline, 7 rats were killed at the 1st, 3rd, 7th, 14th and 28th day after the exposure re- spectively. The expressions of TGF-β1, TβR I and TβR lI in various lung tissues were analyzed by immunohistochemical method and image analysis. Results TGF-β1, TβR I and Ti3R II were widely distributed in lung tissues. The expression of TGF-β1 in the model group was significantly higher than that of the control group ( P 〈 0. 01 ), with the expression reaching the peak at day 7. Compared with those of the control group, the expressions of TβR I and TβR II in model group were significantly higher at the 14th and 28th day, the differences were statistically significant (P 〈 0. 01 ). Conclusion TGFβ-1, TβR I and TβR lI plays an important role in pulmonary fibrosis induced by silica dust.
