奥氮平诱导的小鼠肥胖模型MCP-1水平分析 被引量:2

摘要 目的探讨脂肪因子和重要的炎症趋化刺激因子——单核细胞趋化蛋白1(MCP-1)的血清水平在奥氮平诱导的肥胖中的作用。方法20只雄性小鼠随机分为两组:实验组和对照组,实验组训练小鼠每天自主摄入奥氮平3mg/kg体重。7周后取小鼠血浆并检测血浆MCP-1水平。解剖小鼠并留取附睾脂肪垫及肾周脂肪,计算小鼠腹腔脂肪指数及肥胖指数。结果实验组体重、附睾脂肪垫及肾周脂肪湿重均高于对照组。经计算,腹腔脂肪指数及肥胖指数均高于对照组,可判断为建模成功。实验组血浆MCP-1水平显著高于对照组。结论在奥氮平诱导的肥胖小鼠模型血浆MCP-1水平升高提示炎症可能参与奥氮平诱导的肥胖。 Objective Monocyte chemoattractant protein 1 (MCP--] ) is one of the adipocyto- kines and a inflammatory chemokines stimulating factor. The study is to investigate the role of MCP--1 in olanzapine induced obesity. Methods Mouse were trained to take peanut butter pills voluntarily and then dosed daily with placebo or olanzapine peanut butter pills(3 mg·kg-1·d-1 ). The level of MCP-1 was measured 7 weeks later. Epididymis fat pad and perirenal fat was collected for comparing the obe- sity degree of the two group of mice. Results Compared with control, both the abdominal fat index and the Lee's index used for assessing obesity were higher in olanzapine treatment mice. The levels of MCP -- 1 of olanzapine treatment mice were higher than the control group. Conclusions Inflammation may in-volve in the mechanism of olanzapine induced obesity.
