摘要
目的:研究和探讨CIK细胞杀瘤实验前后细胞表型以及细胞毒性之间的相关性,希望为相关生物学研究提供科学依据。方法:我院恶性肿瘤患者60例于未化疗前取外周血液样本,制备CIK细胞、肿瘤细胞并进行细胞回输,荧光标记法检测细胞表型,液体闪烁法计算细胞毒性,并进行杀瘤前后的比较分析。结果:杀瘤前后淋巴细胞表型经统计学比较具有显著差异(P<0.05),杀瘤后CD3+、CD4+、CD4+/CD8+明显高于杀瘤前,CD8+明显低于杀瘤前。杀瘤前后NK细胞表型经统计学比较具有显著差异(P<0.05),杀瘤后CD3-16+56+明显高于杀瘤前。杀瘤前后CIK杀瘤率经统计学比较具有显著差异(P<0.05),杀瘤后杀瘤率明显高于杀瘤前。杀瘤率与CD3+、CD4+、CD4+/CD8+、NK细胞表型呈正相关(r=0.665,0.736,0.837,P<0.05),与CD8+、CD19呈负相关(r=0.728,0.823,P<0.05)。结论:CIK细胞抗原在肿瘤细胞的杀灭过程中起到了重要的作用,且具有有效性,与淋巴细胞和NK细胞表型呈正相关。
Objective：To study and explore the correlation between cell phenotype and cytotoxic CIK cells kill the tumor before and after the experiment,hoping to provide a scientific basis for the relevant biological research.Methods：60 patients with malignant tumors were taked peripheral blood samples before chemotherapy.To prepare CIK cells,tumor cells and do cell reinfusion.To detect cell phenotype with fluorescence labeling method and cytotoxicity with liquid scintillation method,then to compare and analyze before and after tumor killing.Results： Tumor lymphocyte phenotypes before and after the killing had statistically significant differences（P0.05）,CD3＋,CD4＋,CD4＋/CD8＋after tumor killing were significantly higher than before tumor killing,CD8＋after tumor killing was significantly lower than before tumor killing.NK cell phenotype had statistically significant differences before and after tumor killing（P0.05）,CD316＋56＋ after tumor killing was significantly higher than before tumor killing.CIK kill tumor rate had statistically significant differences before and after tumor killing（P0.05） Tumor killing rate after tumor killing was significantly higher than before tumor killing.Tumor killing rate was positively correlated with CD3＋,CD4＋,CD4＋ / CD8＋ NK cell phenotype（r= 0.665,0.736,0.837,P0.05）,and was negative correlated with CD8＋,CD19（r = 0.728,0.823,P0.05） Conclusion：CIK cell antigens played an important role in tumor cell killing process and had effectiveness,which had positive correlation with the lymphocyte and NK cell phenotype.
