安脑平冲片联合甘露醇注射液对大鼠脑出血后血肿周围MMP-9及TIMP-1表达的影响 被引量:8

Effects of Annaopingchong Pill on the Expression of MMP-9 and TIMP-1 after Intracerebral Hemorrhage in Rats

摘要 目的探讨安脑平冲片联合甘露醇注射液对脑出血后脑水肿的治疗作用及机理。方法在大鼠尾状核注入自体动脉血制作脑出血模型。大鼠随机分为假手术组、模型组、甘露醇组、安脑平冲片组、联合用药组,每组18只,给予相应药物后,分别于脑出血后24 h、48 h、72 h开颅取脑,测定血肿周围脑组织含水量、MMP-9及TIMP-1的表达。结果大鼠脑出血后1~3 d,血肿周围脑组织含水量逐渐增加。模型组与其余几组相比各时间点脑组织含水量均增高;联合用药组脑含水量在72 h时低于甘露醇组与安脑平冲片组。模型组在各测定时间点血肿周围脑组织MMP-9表达强度均较假手术组增加。安脑平冲片组与联合用药组MMP-9的表达强度在24 h、48 h、72 h低于模型组,在24 h和72 h低于甘露醇组(安脑平冲片组24 h)。甘露醇组MMP-9的表达强度在48 h时低于模型组。大鼠脑出血后72 h血肿周围脑组织TIMP-1表达强度升高。安脑平冲片组与联合用药组TIMP-1的表达强度在48 h高于模型组和甘露醇组,联合用药在72 h仍高于模型组和甘露醇组(以上P〈0.05或P〈0.01)。结论安脑平冲片可能通过增加TIMP-1及下调MMP-9的表达水平以减轻脑水肿。其中安脑平冲片和甘露醇在脑出血早期及时联合应用,对于控制脑出血后72 h脑水肿高峰期疗效最佳。 Objective:To investigate the effects of Annaopingchong pill on brain edema with intracerebral hemorrhage and to explore the mechanism,guiding clinical medication.Methods:Intracerebral hemorrhage model in rats was established by injecting autologous arterial blood.The rats were randomly divided into sham-operation group,model group,mannitol group,Annaopingchong pill group,combined treatment group,18 rats in each,to give the corresponding drugs.Takeing the brain specimen,the water content in brain tissue、the expression of MMP-9 and TIMP-1 in intracerebral hemorrhage after 24 h、48 h、72 h were detected.Results:The water content in brain tissue around the hematoma gradually increased in rats with intracerebral hemorrhage after 1-3 days(P0.05).The water contents of the model group were increased compared to the others in each time point;The brain water contents of combination group at 72h bellows the mannitol group(P0.05).The MMP-9 expressions of model group at all time points were up-regulated in the sham-operation group(All P0.01).The MMP-9 expressions of the Annaopingchong pill group and the combined treatment group at 24 h(P0.05),48 h(P0.01),72 h(P0.01) lower than that in the model group,the 24 h and 72 h below the mannitol group(the Annaopingchong pill group at 24 h P0.05,P0.01 for others),The MMP-9 expressions of the Mannitol group in 48h was lower than the model group's(P0.05).The TIMP-1 expressions around hematoma is elevated after intracerebral hemorrhage in rat at 72 h.Annaopingchong pill group and combination group's TIMP-1 expressions at 48 h were higher than that in the model group and the mannitol group(all P0.01),and the combined treatment group's at 72 h was higher than that of the model group and the mannitol group also(P0.05).Conclusion:Annaopingchong pill ameliorates brain edema in rats after intracerebral hemorrhage by inhibiting MMP-9 expressions and activity and up-regulating TIMP-1.The Annaopingchong pill and mannitol on cerebral hemorrhage early combined application for the control of cerebral edema peak at 72 h after intracerebral hemorrhage effect best.
