摘要
目的采用星点设计-效应面法优化健脾益气汤的提取工艺。方法以水煎法提取健脾益气汤中的有效部位,以黄芪甲苷和黄芪多糖的含量为指标,采用星点设计-效应面法考察提取次数、溶媒用量、提取时间3个因素对黄芪甲苷和黄芪多糖的含量影响,优选最佳提取工艺条件。结果最佳提取工艺为加水提取2次,每次1.5 h,溶媒量为8倍。按优化后处方提取的黄芪甲苷和黄芪多糖的含量较高。结论星点设计-效应面法所建立的健脾益气汤提取工艺的数学模型预测性良好。
Objective To optimize the extraction of fianpi yiqi decoction by using central composite design-response surface methodology. Methods The effects of extraction times, material to solvent ratio, and extracting time on the content of Astragaloside IV and Astragalus polysaccharides were investigated. Response surfaces were delineated according to the mathematic models, and optimum formulations were selected. Results All the investigated response variables were highly dependent on the formulation variables. The optimized extraction procedure was as follows: 8 times volume of the weight of the dried material,refluxing 1.5 h for 2 times. The observed values agreed well with model predicted values. Conclusion Central composite design-response surface methodology is successfully used to optimize the extraction offianpi yiql decoction.
