兔关节软骨细胞在纤维蛋白胶中体外培养 被引量:8

摘要 目的 :研究关节软骨细胞在纤维蛋白胶中三维培养时的表现。 方法 :( 1)通过硫酸铵沉淀法从兔血液中提取纤维蛋白原并制备纤维蛋白胶 ;( 2 )在纤维蛋白胶凝成的纤维蛋白中立体培养关节软骨细胞 ,观察其表现并测定生长曲线、倍增时间 ;( 3 )通过透射电镜、AB-PAS染色、 型胶原免疫组化检查等 ,了解细胞生长及软骨基质的分泌情况。结果 :( 1)关节软骨细胞在纤维蛋白中大部分保持卵圆形 ,小部分如贴壁细胞状 ;( 2 )细胞倍增时间在第 1~ 4天为 7.0 d,在第 5~ 7天为 2 .3 d;( 3 )AB-PAS染色 ( +)、 型胶原免疫组化检查 ( +)。结论 :关节软骨细胞能在硫酸铵沉淀法制备的纤维蛋白胶中增殖 。 Objective: To study the behavior of chondrocyte when 3D were cultured in fibrin glue. Methods: (1) Fibrin glue was obtained from rabbit blood through the method of ammonium sulfate precipitation. (2) Chondrocytes cultured in fibrin glue. (3) Histology and electron microscopy were used to study the behavior of chondrocytes cultured in fibrin glue. Results: (1) Most chondrocytes retained their oval shape when cultured in fibrin, a few appeared as they did in monolayer culture. (2) The doubling time was 7.0 days between the first and 4th day, and 2.3 days between the 5th and 7th day. (3) AB PAS stain (+), type Ⅱ collagen immunohistochemical test (+). Conclusion: Chondrocytes grow well and can synthesize cartilage extracellular matrix in fibrin glue.
